背景及目的:去氫表雄固酮(DHEA)是腎上腺皮質製造的主要雄性激素之一,具有調解免疫反應的功能。本研究探討DHEA是否對幼鼠專一性抗體反應具有佐劑功能。 方法:8-10週大的幼鼠免疫注射市售之白喉-非細胞百日咳-破傷風疫苗(DaPT)或以甲醛去活性的百日咳毒素(PT)後,測定血清中抗PT抗體效價及脾臟細胞的細胞激素製造能力。 結果:DaPT疫苗激發很強之抗百日咳毒素(PT)抗體反應,具不受同時注射DHEA及其溶劑(90%乙醇)的影響,接受DaPT疫苗免疫之小鼠脾臟細胞顯著增加干擾素-γ(IFN-γ)、第二介白質(IL-2)及第四介白質(IL-4)的mRNA表現量,而DaPT疫苗與高劑量DHEA(10mg/隻)同時注射則顯著抑制INF-γ及IL-2 mRNA的表現,而IL-4mRNA的表現卻不受影響。去活性PT則激發很弱的抗PT抗體反應,脾臟細胞卻顯著增加第二介白質(IL-2)製造量及第十介白質(IL-10);高劑量DHEA(10mg/隻)同時注射則顯著增強抗PT抗體反應,而脾臟細胞則顯著降低IL-10的製造。 結論:幻鼠免疫注射DaPT疫苗能激發很強的抗PT抗體反應,而IL-4可能是DaPT疫苗免疫小鼠過程中主要的免疫促進因子;DHEA對幼鼠接受百日咳類毒素(PT)免疫注射具有佐劑效應,此效應可能與抑制IL-10製造並回復去活性PT的免疫能力有關。
Background and purpose: Dehydroepiandrosterone (DHEA), one of the predominant androgens secreted by the adrenal cortex, is a potential immunological regulator. In this study, we investigated the possible adjuvant activity of DHEA in young mice. Methods: Young mice (8-12-week-old) were immunized with commercially available diphtheria-acellular pertussis-tetanus (DaPT) vaccine or inactivated pertussis toxin (PT). The anti-PT antibody titer in mouse serum and cytokines secreted by spleen cells were measured. Results: DaPT vaccine induced a high level of anti-PT antibody response, and the antibody response was not affected by either supplementary injection with DHEA or 90% ethanol (solvent for DHEA). The mRNA expression of interferon-γ (IFN-γ), interleukin-2 (IL-2) and interleukin-4 (IL-4) was significantly increased in spleen cells isolated from mice immunized with DaPT vaccine. DaPT vaccine plus high-dose DHEA (10 mg/mouse) significantly suppressed IFN-γ and IL-2 mRNA expression, but IL-4 mRNA expression was unaffected. Immunization with formaldehyde-inactivated PT induced only a mild antibody response. IL-2 and interleukin-10 (IL-10) production was increased in spleen cells isolated from the mice immunized with inactivated PT alone, and inactivated PT plus high-dose DHEA strongly augmented the antibody response against PT. IL-10 production was significantly suppressed in spleen cells isolated from mice immunized with inactivated PT plus high-dose DHEA. Conclusion: The DaPT vaccine induced a strong anti-PT antibody response in young mice, and IL-4 might play a predominant role in this response. DHEA provided an adjuvant effect on the young mice immunized with PT by reducing IL-10 production, which might resume the immunogenicity of PT.
