本研究旨在探討一週運動和單次局部缺血對肌肝醣及GLUT4基因表現(glucose transporter 4 mRNA expression)的影響。本研究以180-190 g Sprague-Dawley rat 16隻作為受試者,分為運動組(n=8)和非運動組(n=8)。運動組老鼠每天進行3小時的游泳運動,持續七天,在第七天運動後自由進食,且在運動後5小時,再以止血帶綁在老鼠右腿上方造成局部缺血1時,之後休息30分鐘進行解剖;非運動組,則是只進行右腿一次的局部缺血。每一組老鼠的右腿作為缺血組,左腿作為非缺血組,因此共有運動缺血組、運動非缺血組、非運動缺血組、非運動非缺血組。本研究測量蹠肌的肌重、腓腸白肌的肌肝醣和GLUT4基因表現。結果發現,缺血與否對肌重、肌肝醣是無影響;運動與否對肌重、肌肝醣也無影響;非運動非缺血組的GLUT4基因表現顯著低於非運動缺血組,運動缺血組的GLUT4基因表現顯著低於非運動缺血組。本研究結論為無論運動或不運動對缺血與否都不影響肌重、肌肝醣,但一週運動會抑制單次局部缺血GLUT4基因表現。
The purpose of this study was to investigate the effect of one-week exercise and acute ischemia on muscle glycogen, and GLUT4 mRNA expression (glucose transporter 4 mRNA expression). Sixteen Sprague-Daweley rats were divided into divided into exercise group (n=8) and un-exercise group (n=8). The exercise group swam 3 hours a day for 1 week. After 5 hour of last bout exercise, the tourniquet was placed around the right upper hindlimb and kept for 1 hour, up to ischemia, and followed 30-min recovery, than the rats sacrificed, whereas the left hind limb serving as control, so that the exercise group subdivided into exercise/ischemia and exercise/un-ischemia. The un-exercise group was applied the ischemia, and followed 30-min recovery, than the rats sacrificed, whereas the left hind limb serving as control, the un-exercise group subdivided into un-exercise/ischemia and un-exercise/un-ischemia. The results of this study showed the exercise and ischemia did not influence the weight of plantaris and the glycogen of gastronemius. The GLUT4 mRNA expression of the exercise/ischemia group was significantly lower than the un-exercise/ischemia group, and the GLUT4 mRNA expression of the un-exercise/un-ischemia group was significantly lower than the un-exercise/ischemia group. The results of this study indicated the weight, glycogen didn't change with the station of ischemia no matter exercising or not exercising, but 1 week exercise can inhibit the GLUT4 mRNA expression.
