應用Nested PCR-DNA定序方法鑑定藿香藥材及其製劑

Identification of Agastache Herb and Its Preparations by Nested PCR and DNA Sequencing Method

摘要


藿香依臺灣中藥典所載為唇形科藿香"Agastache rugosa"(Fisch. et Mey.) O. Kuntze之乾燥地上部,為芳香化濕藥,惟常誤用同為唇形科之另一芳香化濕藥材廣藿香"Pogostemon cablin"(Blanco)Benth.。本研究利用葉綠體基因組中"psbA-trnH"基因間序列區分藿香及廣藿香藥材,並設計藿香專一性引子對,以Nested PCR-DNA定序鑑定製劑中藿香藥材,並可應用於多種製劑。鑑定結果顯示,24件藥材中僅1件檢出藿香,而16件製劑檢體中僅8件檢體檢出藿香,無論藥材或製劑中,藿香檢出率均偏低,顯見誤用情況嚴重。

Medicinal material agastache herb (huo xiang), of which the origin is" Agastache rugosa" (Fisch. et Mey.) O. Kuntze, is often confused with another medicinal material cablin patchouli herb (guang huo xiang), of which the origin is "Pogostemon cablin" (Blanco) Benth. Using universal primers of "psbA-trnH" intergenic sequence, "A. rugosa" could be distinguished with "P. cablin" based on their different PCR product sizes on agarose gel: about 350 and 300 bp respectively. Nested PCR with "A. rugosa"-specific primers could also detect "A. rugosa" in preparations consisting of 7 different kinds of compositions. Surprisingly, "A. rugosa" was only detected in 1 out of 24 raw material samples and 8 out of 16 preparation samples, which reflects the importance of genuine origin identification.
