光、暗及植物賀爾蒙對綠豆發芽期間過氧化酶活性及其異構酶形態之影響

摘要


研究綠豆(phaseolus radialus)在發芽生長過程中過氣化酶之化學性質及光、暗賀爾蒙等處理從其活性變化,並採討其異構酶形態改變。過氧化酶活性隨著綠豆芽生長而增加;在光照環境中活性較高而且不同組織部位所得結果相同。過氧化酶隨著下胚軸之伸長其活性也隨著增加;光、暗、不同環境生長下,其異構酶形態變化不大。利用生化方法純化過氧化酶;在40~60%飽和硫酸銨分割下活性最高;此酵素極耐高溫,在70℃時活性最高且為室溫之3倍左右。它對酸鹼適應範圍甚大;但在pH=6.8時活性較高。利用植物賀爾蒙處理植株後;發現吲嗓乙酸對此酵素活性有抑制現象,而吉貝素及細胞分裂素均有增加活性傾向。檢視見其異構酶結果知IAA可抑制某些異構酶出現,而吉貝素反有促進作用,但細胞分裂素並沒有顯著影響。

The activity of peroxidase was increased as the plant grew. In light enzyme activity was more higher than in dark and that the same as the other tissues. The activity of peroxidase was increased as the hypocotyle grew; In light, dark & various environment growth. The isozyme pat tern was not changed. Enzyme purified by biochemical methods. Enzyme activity was the highest between 40~60% sat. (NH4)2S04 fraction. The enzyme was tolerance to temperature. At 70°C enzyme activity was the highest and triploid of normal temperature. pH range was very broad. but pH=6.8 was the optimal pH. After plant was treated by plant Hormone. IAA inhibited enzyme activity but GA & cytokinin stimulated. Observed the isozyme pattern. IAA depressed the isozyme number, but GA Increased, cytokinin reveal no significant effects.
