Differential Giemsa Staining in Medicago Species

利用吉氏染劑分染法之苜蓿染色體之研究

摘要


利用苜蓿二倍體種Medicago sativa ssp. Caerulea Less., M. sativa ssp. Falcata L. and CADL(M. sativa二倍體試驗系統)三種為材料,進行根端細胞之染色體分染研究。由顯微鏡視察結果得知,M. caerulea及CADL在染色體之末端區,中間區及中節區有明顯的染色世帶(chromosome banding)M. falcata只在染色體之中節區(centromeric region)及第二次縊痕(secondary constriction)處有明顯的染色體帶(C-banding)。此二型染色體帶為苜蓿種之特徵,而此特徵區為安定並能在重複研究中重現。由上面研究結果可預測,利用吉氏染劑分染法(differential giemsa staining)可以正確地規範所有苜蓿種(species)染色體帶之樣式(pattern)。在本文中細胞遺傳對苜蓿育種之應用亦詳加討論。

Diploid species of alfalfa, Medicago sativa ssp. caerulea Less., M. sativa ssp. falcata L. and CADL (an experimental line of cultivated alfalfa at diploid level, M. sativa L.) were used in this study. M. caerulea and CADL had prominent bands at telomeric, intercalary, and probably at centromeric regions. M. falcata had prominent band only at centromeric and secondary constriction regions. These two types of C-banding were characteristic of Medicago species which were con- sistent and reproducible. It is anticipated that precise Giemsa banding patterns should be possible for all or most Medicago species. Their application for alfalfa cytological studies and alfalfa breeding was discussed.
