加減防己黃耆湯對體外培養的人腎小球系膜細胞增殖及產生白細胞介素-8的影響

摘要


目的:觀察了具有解毒、補氣、活血、化濁作用的中藥複方加減防己黃耆湯(以下簡稱FJHQ)對體外培養的人腎小球系膜細胞增殖及產生白細胞介素-8的影響,冀以從細胞生物學水準探討延腎防治慢性腎功能衰竭(chronic renal failure CRF)的作用機制。方法:採用MTT比色法和雙抗夾心ELISA法,觀察FJHQ大鼠血清對體外培養的人腎小球系膜細胞增殖和IL-8生成的影響。結果:LPS可明顯刺激人腎小球系膜細胞增殖和產生IL-8,FJHQ大鼠血清對LPS刺激後人腎小球系膜細胞增殖和產生IL-8有抑制作用,其抑制程度與藥物濃度有一定的量效關係、即隨藥物濃度的增加對系膜細胞增殖和產生IL-8的抑制率也相應增加。結論:加減防己黃耆湯對LPS刺激後腎小球系膜細胞增殖和產生IL-8有明顯的抑制作用,提示加減防己黃耆湯抑制腎系膜細胞增殖和自分泌IL-8是預防腎小球硬化發生發展的重要作用機制之一。

Objective: It was observed that the effect of traditional chinese drugs-Jia Jian Fang Ji Huang Qi Decoction which have the fuction of discharging the turbid, removing toxic materials, tonfing Qi and activating blood on proliferation of human mesangial cells (huMsC)and production of interleukin 8 (IL-8) which is produced by huMsC cultured in vitro. The aim of this study was to approach the mechanism of FJHQ on prevention and treatment of chronic renal failure (CRF) in level of cell biology.Methods: The effects of FJHQ rat serum on proliferation of huMsC and the production of IL-8 by huMsC cultured in vitro were observed. MTT colourmetry and ELISA were used.Results: Proliferation of huMsC and the production of IL-8 can significantly stimulated by LPS in cultured huMsC in vitro, whereas significantly inhibited by FJHQ rat serum and inhibition rat is relative to dosage.Conclusion: It is one of the important mechanism of preventing the occurrence and development of glomerulosclerosis that proliferation of huMsC and the production of IL-8 which produced by MsC were inhibited by FJHQ.
