酸棗仁歸屬於鼠李科(Rhamnaceae)植物酸棗[Ziziphus jujuba Mill. var. spinosa (Bunge) Hu ex H. F. Chou],市售酸棗仁乾燥成熟種子常被外觀相近的滇棗仁(Ziziphus mauritiana Lam.)摻混,因此,有必要加以辨識及區分。本研究以聚合酶連鎖反應(polymerase chain reaction, PCR)鑑別酸棗仁及滇棗仁藥材,成功地建立四個引子對(primer pairs),其中ZjsF1/ZjsR1及ZmITS3F/ZmITS3R引子對可用於鑑別酸棗仁,ZmF1/ZmR1可用於鑑別酸棗仁摻混滇棗仁,而ITS1/ITS4引子對可作為酸棗仁及滇棗仁的共通性基因片段。測試的結果指出聚合酶連鎖反應應用此四個引子對將可快速且有效地鑑別酸棗仁及滇棗仁,以及兩藥材的摻混。
Jujube seeds (Ziziphi spinosae Semen), the dried seeds of Ziziphus jujuba Mill. var. spinosa (Bunge) Hu ex H. F. Chou, are often adulterated with those of Ziziphus mauritiana Lam. for their similarity in appearance. It is therefore important to identify and differentiate the two species. In this study, polymerase chain reactions with four primer pairs (ZjsF1/ZjsR1, ZmITS3F/ZmITS3R, ZmF1/ZmR1 and ITS1/ITS4) were used. Specifically, ZmF1/ZmR1 primer set can detect Ziziphus mauritiana Lam. adulterated in Ziziphus jujuba var. spinosa. ZjsF1/ZjsR1 and ZmITS3F/ ZmITS3R primer sets can identify Ziziphus jujuba var. spinosa. Lastly, ITS1/ITS4 primer set is designed from the conserved gene fragment so can be used to identify both species. Results showed that these four primer sets are useful to identify the two Chinese herbal materials and their authenticity.
