中華被毛孢發酵菌絲體對脂多醣誘發肺部發炎疾病具有保護效果

Protective Effect of Hirsutella sinensis Mycelium on Lipopolysaccharide-induced Acute Lung Injury in Mice

摘要


急性肺損傷為常見的急重症,具有很高盛行率及死亡率,然而,造成急性肺損傷病人全身性發炎反應而導致多重器官衰竭及死亡。中國古籍記載冬蟲夏草(Cordyceps sinensis)為具有潤肺、補腎功效的中藥,中華被毛孢(Hirsutella sinensis)是C. sinensis的無性生殖菌絲體形式。本研究評估中華被毛孢菌絲體H. sinensis mycelium, HSM)萃取物對脂多醣(lipopolysaccharide, LPS)所誘導之小鼠急性肺損傷的保護作用。LPS注射至BALB/c小鼠前,先以腹腔注射中華被毛孢菌絲體萃取物(2mg/g)至BALB/c小鼠,再以氣管注射的方式投予100 μg/50 μl LPS,在施予lipopolysaccharide(LPS,脂多醣類)後24小時將小鼠犧牲,採集每隻小鼠之支氣管肺泡沖洗液(bronchoalveolar lavage fluid, BALF)進行分析。結果發現,以HSM萃取物進行小鼠之前處理可顯著減少BALF中LPS誘導的蛋白質滲出物(protein exudate)。此外,與LPS組小鼠相比,LPS+HSM提取物組BALF中總細胞、白血球(CD45)、吞噬細胞(CD45^+/CD11b^+)和嗜中性球(CD45^+/Ly6G^+)的數量顯著減少。這些發現顯示HSM萃取物通過減輕肺部炎症而可預防LPS誘導的急性肺損傷。

Acute lung injury (ALI) is common with a high mortality rate. Systemic inflammation is implicated in the progression of acute lung injury leading to multiple organ failure and death. Cordyceps sinensis (C. sinensis) has been used as a traditional Chinese medicine to invigorate the lung and nourish the kidney. Hirsutella sinensis is the anamorphic, mycelial form of C. sinensis. In the present study, we investigated the protective effects of Hirsutella sinensis mycelium (HSM) extract on lipopolysaccharide (LPS)-induced acute lung injury in mice. HSM extract (2 mg/g) was intraperitoneally injected into BALB/c mice 0.5 hour before LPS injection, and then 100 μg/50 μl LPS was administered by tracheal injection. All mice were sacrificed 24 h after LPS treatment, and bronchoalveolar lavage fluid (BALF) was collected from each mouse for subsequent analyses. Results showed that pretreatment of mice with HSM extract significantly reduced protein exudate induced by LPS in BALF. In addition, the numbers of total cells, white blood cells (CD45), phagocytes (CD45^+/CD11b^+), and neutrophils (CD45^+/Ly6G^+) in BALF were significantly decreased in the LPS + HSM extract group compared to the LPS group of mice. These findings suggest that HSM extract prevents LPS-induced acute lung injury by attenuating lung inflammation.
