GNB-14電腦密碼鑑定系統可有效地輔助常見革蘭氏陰性桿菌的鑑定

Evaluation of GNB-14 Computer-Coding System for Identification of Common Gram-negative Bacilli

摘要


革蘭氏陰性桿菌為臨床或食品微生物檢驗室常見的分離菌,這些菌的傳統鑑定特性約有50種,為了減少鑑定發生錯誤,吾等開發了GNB-14電腦密碼系統作為輔助革蘭氏陰性桿菌的鑑定。本研究將利用常見的24個嗜氧性與兼性厭氧性革蘭氏陰性菌菌種,共141株,包括51株葡萄糖發酵性菌和90株非發酵性菌評估GNB-14電腦密碼鑑定系統輔助鑑定的準確性。測試時,以接種針挑取任一測試株的菌落接種八種生化試驗培養基:(1)triple sugar iron agar(三糖鐵瓊脂,TSIA);(2)Simmon's citrate agar(西蒙式檸檬酸鹽瓊脂,CIT);(3)Christensen's urea agar(克氏尿素瓊脂,URE);(4)sulfide-indole-motility medium(硫化物-吲哚-運動性培養基,SIM);(5)Voges-Proskauer semi-solid medium(歐普氏試驗培養基,VP);(6)Moeller's ornithine decarboxylase medium(默勒氏鳥氨酸脫羧酶培養基,ORN);(7)Moeller's arginine dihydrolase medium(默勒氏精氨酸雙水解酶培養基,ARG);(8)Moeller's lysine decarboxylase medium(默勒氏賴氨酸脫羧酶培養基,LYS)以及操作oxidase(氧化酶)試驗。接種後,將此些試驗管置於35℃一般培養箱培養18~20小時後判讀,可獲得14種反應特性,將其轉換為5個數字的菌碼,再查閱GNB-14電腦密碼鑑定系統本文,結果指出此系統輔助常見葡萄糖發酵性菌和非發酵性革蘭氏陰性桿菌的準確性高達99.3%(140/141)。總之,GNB-14電腦密碼鑑定系統可有效地輔助臨床或食品微生物檢驗室鑑定常見的嗜氧性或兼性厭氧性革蘭氏陰性桿菌。

Gram-negative bacilli are commonly isolated in clinical or food microbiology laboratories, and more than 50 characteristics are used for their identification. To minimize identification errors, we have developed the GNB-14 computercoding system. In this study, the accuracy of this system was evaluated using 141 strains of commonly encountered aerobic and facultatively anaerobic Gram-negative bacilli, including 51 strains of glucose fermenters and 90 strains of non-fermenters. Each colony of a strain to be tested was inoculated in the following 8 biochemical test media: (1) triple sugar iron agar; (2) Simmon's citrate agar; (3) Christensen's urea agar; (4) sulfide-indole-motility medium; (5) Voges-Proskauer semi-solid medium; (6) Moeller's ornithine decarboxylase medium; (7) Moeller's arginine dihydrolase medium; (8) Moeller's lysine decarboxylase medium. Also, the oxidase test was performed. After incubation of the inoculated media at 35℃ for 18-20 hours, 14 characteristics were obtained. These characteristics were then converted into 5-digit codes. Results showed that GNB-14 correctly identified 140 of 141 (99.3%) glucose-fermenting and non-fermenting Gram-negative bacilli. In conclusion, the GNB-14 computer-coding system is effective in aiding the identification of common Gram-negative bacilli in clinical or food microbiology laboratories.
