評估MicroFast和3M Petrifilm在臺灣手搖飲料中的微生物檢測效能

Evaluation of the Effectiveness of MicroFast and 3M Petrifilm in Microbial Testing of Hand-Shaken Drinks in Taiwan

摘要


臺灣食品藥物管理署(TFDA)公布許多檢測食品和飲料中微生物的標準化方法,這些微生物括生菌數、酵母菌與黴菌、腸桿菌科、大腸桿菌群與大腸桿菌等。幾種常見廠牌的簡便檢驗套組,如MicroFast(Meizheng Bio-tech Co., Ltd., PRC)和3M Petrifilm(3M Co., Ltd., USA)等,均通過AOAC國際組織認證且已經上市。在本研究中,吾等檢查53種手搖飲料,以TFDA或FDA/USA方法評估這些套組的偵測效能。用4種不同方法檢測出不合格/陽性檢體的百分比和件數如下:(i)生菌數>1×10^4 CFU/mL:TFDA 33.9%(18/53)、MicroFast AC 32.1%(17/53)與3M Petrifilm AC 27.2%(12/44);(ii)酵母菌與黴菌:TFDA 97.7%(42/43)、MicroFast YM 95.3%(41/43)與3M Petrifilm YM 96.8%(30/31);(iii)腸桿菌科:TFDA 95.6%(43/45)、MicroFast EB 88.9%(40/45)與3M Petrifilm EB 73.7%(14/19);(iv)大腸桿菌群:FDA/USA 77.6%(41/53)、MicroFast EC 66.0%(35/53)與3M Petrifilm EC 54.5%(24/44);以及(v)大腸桿菌(Escherichia coli):均未檢出。這些結果指出,儘管MicroFast和3M Petrifilm的各種檢驗套組容易操作,但其等偵測生菌數、腸桿菌科與大腸桿菌群的效能仍有待提高,其中MicroFast系統對生菌數、腸桿菌科與大腸桿菌群之偽陰性率分別為1.8%、6.7%與11.6%,而3M Petrifilm系統高達6.7%、21.9%與23.1%,此值得使用者加以注意。由於市場上手搖飲料的微生物汙染率甚高,吾等敦促衛生主管部門定期監測,以確保其安全性。

The Taiwan Food and Drug Administration (TFDA) has published standardized methods for detection of microorganisms in foods and drinks. These microorganisms may include aerobic plate count, yeast and mold count, Enterobacteriaceae, coliform bacteria, and Escherichia coli. Several test kits such as MicroFast (Meizheng Bio-tech Co., Ltd., PRC) and 3M Petrifilm (3M Co., USA), which have been accredited by the AOAC International, are commercially available. In this study, we examined 53 hand-shaken drinks to assess the detection performance of these kits by the TFDA method. Percentage and number of samples tested positive by the 4 different methods are as follows: (i) aerobic bacteria > 1×10^4 CFU/mL: TFDA 33.9% (18/53), MicroFast AC 32.1% (17/53), and 3M Petrifilm AC 27.2% (12/44); (ii) yeast and mold: TFDA 97.7% (42/43), MicroFast YM 95.3% (41/43), and 3M Petrifilm YM 96.8% (30/31); (iii) Enterobacteriaceae: TFDA 95.6% (43/45), MicroFast EB 88.9% (40/45), and 3M Petrifilm EB 73.6% (14/19); (iv) coliform bact eria: FDA/USA 77.6% (41/53), MicroFast EC 66.0% (35/53), and 3M Petrifilm EC 54.5% (24/44); and (v) Eschereichia coli: not detected by any of the 3 methods. These results indicated that although both MicroFast and 3M Petrifilm methods are easy to perform, their effectiveness on the detection of aerobic plate count, Enterobacteriaceae, and coliform bacteria remains to be improved. The false-negative rates of MicroFast system, Enterobacteriaceae and E. coli were 1.8%, 6.7% and 11.6% respectively, while 3M Petrifilm system was as high as 6.7%, 21.9% and 23.1%, which should be noted by users. As there is a high rate of microbial contamination in hand-shaken drinks on the market, we urge the health authorities to regularly monitor them in order to ensure their safety.
