Analysis of Food Microbiological Proficiency Test Results

摘要


能力試驗是將模擬檢體結果與其他實驗室結果進行交叉比對的平臺,透過整體的評估,能夠判斷參與實驗室內部的分析能力。本研究分析2018年至2022年331家實驗室的11項食品微生物能力試驗計畫結果,除了分析能力試驗對參加實驗室檢測效益外,還調查微生物試驗數據異常值和不滿意原因。結果指出,對照檢測結果可以發現檢體測試和資料分析中的錯誤,並為參與實驗室提出適當的糾正措施。導致微生物分析失敗的原因可能是檢體處理或測試時和結果分析中發生錯誤。分析前的階段,常見的錯誤主要是由於檢體儲存溫度和儲存時間不當所造成的;分析中的階段,失敗的原因很多,包括檢體的前處理或測試方法不當、檢體混淆、稀釋錯誤、培養基或試劑品質不良、菌落計數錯誤、未執行陰性和陽性對照、環境干擾和分析儀故障;而在分析後的階段,失敗的原因可能是計算方法錯誤和結果記錄謬誤。本研究對這些因素提出避免錯誤的實用解決方案,以作為提高微生物實驗室測試能力的指引。

Proficiency testing is a platform for cross-checking simulated specimen results with those of other laboratories. The results are then used to evaluate analytical capabilities of participating laboratories. In this study, the results of 11 food microbiology proficiency tests of 331 laboratories from 2018 to 2022 were analyzed. In addition to analyzing the benefits of proficiency testing, causes for failing microbiological tests were also investigated. Results showed that cross-referencing of test results could reveal errors in sample testing and data analysis and suggest appropriate corrective measures for participating laboratories. Causes for failing a microbiological analysis could be due to errors in specimen processing, or testing and result analysis. Errors that commonly occurred at the pre-analysis step were mainly due to inappropriate sample storage temperature and duration. Many causes for failure at the analysis step were found, including inappropriate sample pre-treatment or test methods, mi xed samples, dilution errors, poor quality of media or reagents, colony counting errors, inappropriate negative and positive controls, environmental disturbances, and machine malfunctions. In the post-analysis step, failures could be due to wrong computation methods and mistakes in result recording. Results of this study could serve as a guide for improvement in testing capability of microbiology laboratories.
