用非黏蛋白(mucin)培養基量產次世代益生菌Akkermansia muciniphila

Mass Production of Next-Generation Probiotic Akkermansia muciniphila with a Non-mucin Culture Medium

摘要

Akkermansia muciniphila (Akk) is one of next-generation probiotics and has been shown to have the potential to prevent and ameliorate health problems such as obesity, diabetes, metabolic syndrome, and inflammation. As AKK is a strict anaerobe Gram-negative bacterium, its mass production has been very challenging. Since native AKK uses mucin in the intestine for growth, it is traditionally cultured in medium with mucin. In this study, we compared the growth of an AKK strain, which we had isolated from a healthy person, to that of the reference strain (BAA-835) in a 50-L fermenter with a non-mucin, phosphate-buffered culture medium containing glucose, yeast extract, N-acetyl glucosamine, and L-cysteine as carbon and protein sources. After 42 hours of cultivation, the bacterial count in the fermentation broth of the isolated strain (4.1×10^9 CFU/mL) was higher than that of BAA-835 (2.3× 10^9 CFU/mL). However, the bacterial count in the pellet of BAA-835 (9.1×10^(10) CFU/mL) was higher than that of the isolated strain (1.7×10^(10) CFU/mL). After mixing with protectants, the freeze-dried bacterial count of the isolated strain (5.9×10^(10) CFU/g) was higher than that of BAA-835 (2.2×10^(10) CFU/g). After one month of storage at 30℃ , the bacterial count of BAA-835 in the freeze-dried powder was decreased by 73% (to 6.0×10^9 CFU/g), while that of the isolated strain was decreased by 94% (to 3.3×10^9 CFU/g). This is the first reports of AKK mass production with a non-mucin culture medium.
