CMP^(TM)efSwab Buffered Glycerol Saline(efSwab BGS)和CMP^(TM)efSwab Cary-Blair broth(efSwab CB)是糞便檢體的非營養性液體輸送培養基,兩者可與Kiestra InoqulA(BD, USA)或WASP(Copan, Italy)自動微生物接種系統相容。本研究評估此兩種裝置對志賀氏菌種的保存效果。初步測試使用含有沙門氏菌和大腸桿菌的模擬檢體,在檢體中添加高或低量的Shigella sonnei(宋內氏志賀氏菌)(高:500 CFU;低:50 CFU)或S. flexneri(福氏志賀氏菌)(高:600 CFU;低:60 CFU)。結果顯示,對含有大量志賀菌的檢體,efSwab BGS和efSwab CB都能在4℃或25℃下保存S. sonnei和S. flexneri達24小時。對於含有少量志賀氏菌的檢體,efSwab CB在4℃或25℃下可以保存24小時,但efSwab BGS保存S. sonnei於4℃下可以保存24小時,25℃下保存6小時,而S. flexneri在任何溫度下都只能保存6小時。為了評估兩種裝置保存糞便中志賀氏菌的有效性,我們使用了在0.45% NaCl溶液中添加高量(1,100 CFU的S. sonnei和600 CFU的S. flexneri)或低量(110 CFU的S. sonnei和60 CFU的S. flexneri)志賀氏菌的糞便檢體。結果指出對於含有大量志賀菌的檢體,efSwab CB和efSwab BGS都能在4℃或25℃下保存S. sonnei和S. flexneri達24小時。對於含有少量志賀氏菌的檢體,efSwab CB和efSwab BGS均可在4℃和25℃下保存S. sonnei達24小時,但兩種培養基皆無法在25℃下保存S. flexneri達24小時。這些結果顯示,如果檢體在採檢後當天運送,efSwab CB和efSwab BGS都可以使用;如果檢體需要存放一段較長時間(例如過夜)才接種至平板,則建議將efSwab CB或efSwab BGS以低溫(置放冰寶)運送,並保存在4℃冰箱。
CMP^(TM) efSwab Buffered Glycerol Saline (efSwab BGS) and CMP^(TM) efSwab Cary-Blair broth (efSwab CB) are non-nutritive liquid transport media for fecal samples and are compatible with automated microbial inoculation systems such as Kiestra InoqulA (BD, USA) or WASP (Copan, Italy). In this study, we evaluated their effectiveness in the preservation of Shigella spp. For initial testing, simulation specimens containing Salmonella sp. and Escherichia coli spiked with high or low amounts of S. sonnei (high: 500 CFU; low: 50 CFU) or S. flexneri (high:600 CFU; low: 60 CFU) were used. Results showed that both efSwab BGS and efSwab CB could preserve both S. sonnei and S. flexneri up to 24 hours at 4℃ or 25℃ for the samples with high amounts of Shigella spp. For the samples with low amounts of Shigella spp, efSwab CB can preserve them up to 24 hours at 4℃ or 25℃ , while efSwab BGS can preserve S. sonnei up to 24 hours at 4℃ and 6 hours at 25℃ . However, efSwab BGS could only preserve S. flexneri up to 6 hours at either temperature. To evaluate their effectiveness in preserving Shigella spp in stool, fecal samples in 0.45% NaCl solution spiked with high (1,100 CFU for S. sonnei and 600 CFU for S. flexneri) or low (110 CFU for S. sonnei and 60 CFU for S. flexneri) amounts of Shigella spp were used. For samples with high amounts of Shigella spp, both efSwab CB and efSwab BGS could preserve both S. sonnei and S. flexneri at 4℃ or 25℃ up to 24 hours. For samples with low amounts of Shigella spp, both efSwab CB and efSwab BGS can preserve S. sonnei up to 24 hours at both 4℃ and 25℃ but failed to preserve S. flexneri at 25℃ for 24 hours. These results suggest that both efSwab CB and efSwab BGS can be used if the samples are to be transported promptly and that efSwab CB or efSwab BGS be shipped at low temperatures (in ice packs) and stored in a 4℃ refrigeratorif the samples need to be stored for an extended period (e.g. overnight) before being inoculated to the plates.
