去除皮下脂肪與瘦體素補充處理對肥胖小鼠胰島素阻抗及內臟脂肪之影響

Effects of Subcutaneous Fat Removal and Leptin Repletion on Insulin Resistance and Visceral Fat Accumulation in Dietary-Obese Mice

摘要


去除皮下脂肪會造成個體的內臟脂肪增加,而內臟脂肪的增加已知與胰島素阻抗相關之代謝性疾病的發生有關。此外瘦體素已知有降低高血糖、高血脂及高血胰島素等作用。本實驗以高脂飼料誘發肥胖小鼠為材料,進行探討動物在經過去除皮下脂肪與瘦體素補充處理後,能否改善其胰島素阻抗情形及降低內臟脂肪堆積。實驗結果顯示去除皮下脂肪的肥胖小鼠會有相對較多的內臟脂肪累積,較差的胰島素活性及較高的血脂濃度。瘦體素補充處理並不影響去除皮下脂肪的肥胖小鼠的內臟脂肪累積量及胰島素作用。本實驗的結果指出肥胖小鼠在去除皮下脂肪後反而會惡化其胰島素阻抗情形。去除皮下脂肪後的內臟脂肪的回饋性增加現象,可能與瘦體素的作用無關。

The removal of subcutaneous fat could cause a compensatory accumulation of visceral fat that closely related to the manifestation of insulin resistance related metabolic diseases. Moreover, leptin repletion corrects certain disorders, like hyperglycemia, hyperinsulinemia and hyperlipidemia. We investigated if visceral fat accumulated to aggravate insulin resistance in dietary obese mice which had received subcutaneous fat removal. Exogenous leptin treatment on the changes of insulin resistance and visceral fat accumulation in those lipectomized obese mice was also examined. The results showed that lipectomized obese mice had more visceral fat, higher blood lipid levels, and reduced insulin sensitivity. However, leptin repletion did not have significant improvements on reducing visceral fat deposition nor insulin resistance in those lipectomized obese mice. Our data indicate that the removal of subcutaneous fat could led to visceral fat accumulation and the aggravation of insulin resistance. Moreover, leptin may not participate in lipectomy induced compensatory visceral fat deposition.
