人類臍帶間質幹細胞外泌體美白能力之研究

A Study on The Antioxidant and Whitening Capacity of Exosomes from Human Umbilical Cord Mesenchymal Stem Cells

摘要


外泌體是近年來興起的新興材料,其功能之一是作為細胞間溝通的橋樑。有研究指出不同細胞的外泌體有各自的效果,而人類臍帶間充質幹細胞外泌體具有抗發炎、促進皮膚再生、抗光老化、抗氧化等潛在功能。關於外泌體美白相關研究數據甚少,本研究擬探討人類臍帶間充質幹細胞外泌體之美白作用與作用機制,利用測試細胞外蘑菇酪胺酸酶活性、B16F10存活率、細胞內酪胺酸酶活性與黑色素含量、ROS含量、西方墨點法進行評估。結果顯示人類臍帶間充質幹細胞外泌體對於蘑菇酪胺酸酶活性具抑制效果,最高達33.88±6.17%,且對於B16F10細胞無毒性,能有效降低黑色素含量為對照組的75.31±1.12%及降低細胞內酪胺酸酶活性為對照組的77.80±4.32%,有效降低B16F10細胞內ROS含量為對照組的61.06±3.30%。在西方墨點法的顯影圖中可以觀察到美白相關蛋白質包括Tyrosinase、TRP-1、TRP-2和MITF隨著hucMSC-Exos作用濃度增加,條帶有變淺,可以初步判斷hucMSC-Exos是透過影響上述四種蛋白來達到抑制黑色素生成之效果。綜上述結果,未來可將人類臍帶間充質幹細胞外泌體作為美白保養品的原料開發,並深入探討可以應用的相關美白產品劑型。

Exosomes are emerging materials that have emerged in recent years, and one of their functions is to serve as a bridge for intercellular communication. Some studies have pointed out that exosomes from different cells have their own effects, and human umbilical cord mesenchymal stem cell exosomes (hucMSC-Exos) have potential functions such as anti-inflammation, promotion of skin regeneration, anti-photoaging, anti-oxidative stress, etc. There are few research data on exosome whitening, so this study explores the whitening ability and possible mechanism of hucMSC-Exos, the inhibitory ability of extracellular mushroom tyrosinase activity, B16F10 survival rate test, melanin content measurement, intracellular tyrosinase activity inhibitory ability, ROS content test and Western blotting for evaluation. The results showed that hucMSC-Exos had an inhibitory effect on mushroom tyrosinase activity, up to 33.88±6.17%, and they had no toxicity to B16F10 cells;moreover, they could effectively reduce melanin content to 75.31 ± 1.12% of the control group and reduced intracellular tyrosinase activity. The residual enzyme activity was 77.80 ± 4.32% of the control group.The effective reduction of ROS content in B16F10 cells was 61.06 ± 3.30% of the control group. It can be observed in the western blot result that the bands of whitening-related proteins, including tyrosinase,TRP-1,TRP-2 and MITF become lighter as the concentration of hucMSC-Exos increases. It can be preliminarily judged that hucMSC-Exos affect the above four proteins to achieve the effect of inhibiting melanin production. Based on the above results, human umbilical cord stem cell exosomes can be used as raw materials for whitening skin care products in the future, and the formulations of relevant whitening products that can be applied will be discussed in depth.
