臺灣家蠶核多角體病毒去氧核醣核酸之分析與基因庫之建立

ANALYSIS OF DNA AND GENOMIC BANK CONSTRUCTION OF NUCLEAR POLYHEDROSIS VIRUS OF THE SILKWORM, BOMBYX MORI, ISOLATED IN TAIWAN

摘要


自臺灣罹病家蠶體中分離及純化之家蠶核多角體病毒(BmNPV),所含之DNA,以不同種限制酶切割後,以洋菜膠電泳分析,估算其DNA大小,得知該病毒DNA之分子量約為120 kb左右。將純化之BmNPV DNA,利用加熱方式,觀察其雙股DNA,在裂解成為單股DNA過程中,吸光值在波長260 nm之變化。相對吸光值隨溫度之增加,呈現典型之S型上升曲線,由此曲線測得Tm (melting temperature)約為71.3 C,換算得GC總含量約為42.5%,而其增色性(hyperchromicity)約為37.02%,此病毒具有一多角體蛋白基因(polyhedrin gene),以高效率合成病毒之多角體蛋白,適合作為遺傳工程上異源蛋白合成時之載體(vector)。為尋找此基因,首先將完整之病毒DNA,以限制酶Sau 3 AI切割,分別揷到噬菌體載體EMBL3內,藉不同噬菌體來保存所有病毒DNA,此不同重組噬菌體卽組成BmNPV基因庫(genomic bank)。以本實驗所用噬菌體載體之容量與BmNPV DNA之分子量計算得知,218株重組噬菌體中涵蓋病毒所有基因之機率遠超過99.99%,故可從中篩選多角體蛋白基因。以日本病蠶體中分離出之BmNPV多角體蛋白基因為DNA探子,在低鹽高溫沖洗情形下,自上述臺灣BmNPV基因庫共篩選到4株重組噬菌體,可能含有多角體蛋白基因,其中一株經分析顯示極可能含有完整之多角體蛋白基因,將可作為多角體蛋白基因進一步之選殖(cloning),並提供表現載體構築之材料。

The total DNA of nuclear polyhedrosis virus of the silkworm, Bombyx mori, (BmNPV) isolated in Taiwan was estimated to be ca. 120 kb as determined from the size of the DNA fragments generated by restriction endonuclease digestion. The GC content of BmNPV DNA was ca. 42.5% as calculated from its melting temperature of 71.3 C, its hyperchromicity being ca. 37.02%. The polyhedrin gene of BmNPV (Taiwan) was studied by constructing a genomic bank using phage EMBL3 after digesting with the restriction enzyme Sau 3AI. Among 218 recombinant phages obtained from hybridization with BmNPV (Japan) polyhedrin gene probe, 4 of them showed signal, indicating that they contain the polyhedrin gene. Single plaque formed by one recombinant phage as hybridized repeatedly with the probe was cloned for further study of the polyhedrin gene structure.
