鐵依賴性細胞死亡(ferroptosis)是近年來發現的一種調控性細胞死亡方式,迥異於其他細胞死亡方式,且在多種疾病皆扮演重要角色,其中包括缺血性腦中風。Ferroptosis之機轉主要牽涉三大部分,分別為鐵質代謝異常、胺基酸代謝異常、及脂質過氧化。已有許多基礎研究證實ferroptosis對於缺血性腦細胞損傷有重要影響。此篇綜論的重點在於闡述ferroptosis三大機轉在缺血性腦中風之證據,並介紹ferroptosis與缺血性腦中風之神經發炎的關係。至於ferroptosis指標在腦中風病患之角色,以及抑制ferroptosis在腦中風治療上的成效,皆尚待進一步的臨床研究證實。
Ferroptosis is a recently identified programmed cell death, which is different from other types of cell death. It has been proven to play pivotal roles in various kinds of diseases, including ischemic stroke. The mechanism of ferroptosis involves three major parts, which are abnormal metabolism in iron and amino acid, and lipid peroxidation. Growing evidence from basic experiments has shown the important influence of ferroptosis on the cell death in ischemic stroke. The aim of this review is to summarize the evidence of the three major parts of ferroptosis in ischemic stroke, and to introduce the relationship between ferroptosis and neuroinflammation. Further study is worthy to clarify the roles of ferroptosis biomarkers and the efficacy of inhibiting ferroptosis in patients with ischemic stroke.
