蛹蟲草萃取物對於化療小鼠心肌損傷與骨質疏鬆的保護作用

Protective effect of Cordyceps militaris extract on myocardial injury and osteoporosis in chemotherapy mice

摘要


癌症患者接受化療容易引起心毒性與骨質疏鬆,過去研究認為蛹蟲草(Cordyceps militaris)具有抗腫瘤、免疫調節等保健功效,而蛹蟲草是否有緩解化療造成不良副作用的功效則有待探討。為此,本研究主要探討蛹蟲草萃取物對於艾黴素(adriamycin)化療處理的H9c2心肌細胞以及ICR雌鼠是否具有心肌保護以及緩解骨質疏鬆的功效。本實驗利用DPPH檢測,結果顯示蛹蟲草萃取物具有極佳的抗氧化能力;使用MTT分析,結果顯示蛹蟲草萃取物對於艾黴素誘發心肌細胞損傷具有極佳的保護作用;利用酵素結合免疫吸附(ELISA)檢視,結果發現蛹蟲草萃取物可以提升化療小鼠血清中IgG與IgA表現,但抑制IgE表現;利用免疫組織化學染色以及西方墨點法測定,結果顯示蛹蟲草萃取物具有提升化療小鼠心肌組織抗氧化壓力蛋白質SOD2表現,但是抑制發炎蛋白質TNF-α表現;利用小鼠脛骨電腦斷層攝影,結果顯示蛹蟲草萃取物具有增加化療小鼠脛骨組織骨密度效果。綜合上述,蛹蟲草萃取物對於化療小鼠引起心毒性與骨質疏鬆確實具有保護以及緩解的功效。

Chemotherapy in cancer patients is prone to cause cardiotoxicity and osteoporosis. In the past, it has been reported that Cordyceps militaris has anti-tumor, immune regulation and other health benefits. Whether Cordyceps militaris has the effect of alleviating adverse side effects caused by chemotherapy remains to be explored. Therefore, this study mainly explored whether the addition of Cordyceps militaris extract had cardioprotective and osteoporotic effects on H9c2 cardiomyocytes and ICR female mice treated with adriamycin chemotherapy. In this experiment, DPPH detection was used to show that the Cordyceps militaris extract of extract had excellent antioxidant capacity. MTT assay was used to show that the Cordyceps militaris extract has an excellent protective effect on damaged myocardial cell by adriamycin. Enzyme-linked immunosorbent assay (ELISA) was used to show that the Cordyceps militaris extract can enhance IgG and IgA expressions, but inhibit IgE expression in the serum of chemotherapy mice. Immunohistochemical staining and Western blotting methods was used to show that the Cordyceps militaris extract can enhance antioxidant-related SOD2 protein, but inhibited inflammatory-related TNF-α protein expressions in myocardial tissue of chemotherapy mice. Computer tomography was used to show that the Cordyceps militaris extract has the effect of increasing the bone density of tibia tissue of chemotherapy mice. Based on the above results, the Cordyceps militaris extract should have protective and relieving effects on cardiotoxicity and osteoporosis induced by chemotherapy in mice.
