Lacticaseibacillus paracasei Yb(副乾酪乳桿菌Yb)BCRC 910415為長期應用於發酵乳中之益生菌株,本研究藉由餵食小鼠(BALB/c)探討其免疫調節的能力,試驗以人工管餵方式,持續餵飼12週不同劑量的L. paracasei Yb,並以卵白蛋白(ovalbumin, OVA)致敏。實驗發現,餵飼L. paracasei Yb,不會影響先天性免疫反應的單核球(monocyte)及嗜中性白血球(neutrophil)之吞噬活性,及脾臟中自然殺手細胞(natural killer cells)的毒殺能力。每隻小鼠每日餵飼中劑量組L. paracasei Yb(1.5 × 10^7 CFU/mouse/day)及高劑量組(1.5× 10^8 CFU/mouse/day)可調降血清中的免疫球蛋白(immunoglobulin, Ig)G及IgE的濃度;高劑量組並可顯著降低卵白蛋白特異性免疫球蛋白(OVA-specific IgE)含量(P < 0.05)。脾臟細胞以裂殖素concanavalin A(Con A)及OVA,刺激其中的T輔助細胞(T helper cell, T_H cell),測定其第一型T輔助細胞(Type 1 T helper, T_H1 cell)之細胞激素IFN-γ,及T_H2細胞之細胞激素IL-4、IL-5、IL-13分泌量,並以T_H1細胞激素/T_H2細胞激素的比值,作為T_H細胞分化趨向的指標,餵飼L. paracasei Yb具有促使往T_H1細胞分化的趨向。由調降血清中免疫球蛋白的濃度,及促使T_H1細胞的分化趨向,推測L. paracasei Yb具有減緩OVA誘發的過敏反應,及免疫調節的能力。
Lacticaseibacillus paracasei Yb (BCRC 910415) is a probiotic strain long-term used in fermented milk drinks. This study evaluates the immune regulatory function in L. paracasei Yb. In the experiment, BALB/c mice were gavaged with different doses of L. paracasei Yb for 12 weeks and sensitized with the ovalbumin (OVA). Results showed that feeding L. paracasei Yb did not affect the phagocytosis of monocytes and neutrophils in the innate immune response and the cytotoxic activity of natural killer cells in the spleen. Mice administered with medium (1.5 × 10^7 CFU/mouse/day) and high (1.5 × 10^8 CFU/mouse/day) doses of L. paracasei Yb showed a reduction of IgG and IgE in mouse serum. The high dose L. paracasei Yb intervention significantly reduces the OVA-specific IgE content (P < 0.05). Splenocytes were stimulated with concanavalin A and OVA separately to evaluate the concentration of IFN-γ, IL-4, IL-5, and IL-13 secreted from T helper cells. We found that mice treated with L. paracasei Yb elevated the ratio of T_H1 / T_H2 cytokines. These results indicated L. paracasei Yb reduced specific immunoglobulin concentration and enhanced T_H1 cells polarization. Thus, it can be concluded that administration of L. paracasei Yb might attenuate allergic symptoms and regulate the immune function.
