薏仁中的非澱粉多醣已在許多生理學研究中顯示出其生物活性。本研究於臺灣薏仁中發現阿拉伯半乳聚醣,其為高等植物中已知具生物活性的多醣。阿拉伯半乳聚醣為非消化水溶性多醣的部分組成。從麩皮中分離出的阿拉伯半乳聚醣其單醣組成為阿拉伯糖:半乳糖:葡萄糖:葡萄糖醛酸,莫耳數比為20:25:1:3。半乳糖的醣苷鍵結為3,6-Galp、3-Galp、3,4,6-Galp、6-Galp,阿拉伯糖的醣苷鍵結主要為T-Araf、5-Araf。純化的阿拉伯半乳聚醣作為標準品,建立β-Gal-Yariv呈色法來定量台灣薏仁中的阿拉伯半乳聚醣含量。結果顯示,糙薏仁的含量範圍為0.24-0.27 mg/g,精白薏仁的含量範圍0.15-0.22 mg/g。阿拉伯半乳聚醣在麩皮、胚、胚乳的分佈比例分別為27.32-32.53%、29.75-45.50%、9.52-51.44%。使用市售檢測套組分析薏仁,發現僅含有微量的果寡醣、果聚醣、β-(1,3;1,4)-D-葡聚醣。本研究建立的阿拉伯半乳聚醣定量方法可應用於薏仁及薏仁多醣產品的品質管制。
The non-starch polysaccharides in coix kernels have shown its bioactivity attributes in many physiological studies. This research revealed the molecular characteristics of arabinogalactan, a known bioactive polysaccharide in high plants, in grains of coix cultivars grown in Taiwan. The arabinogalactan was a component of indigestible water-soluble polysaccharides. The monosaccharide composition of isolated arabinogalactan from bran is arabinose: galactose: glucose: glucuronic acid in the molar ratio of 20 : 25 : 1 : 3. The glycosidic linkages of galactose are 3,6-Galp, 3-Galp, 3,4,6-Galp, 6-Galp, and arabinose, primarily T-Araf and 5-Araf. This isolated arabinogalactan was used as a reference standard to quantify the content of arabinogalactan in kernels of coix cultivars grown in Taiwan by a developed β-Gal-Yariv colorimetry. The results showed that the content was in the range of 0.24-0.27 mg/g for whole grain (brown adlay) and 0.15-0.22 mg/g for the polished kernel (white adlay). The ratios of distribution of arabinogalactan in bran, embryo, and endosperm are 27.32-32.53%, 29.75-45.50%, and 9.52-51.44%. The coix selected only contained a trace amount of fructooligosaccharides and fructans, and β-(1,3;1,4)-D-glucans measured by using commercial assay kits. The quantification method for arabinogalactan developed in this study can be applied to the quality control of coix kernel and polysaccharide products.
