洋蔥汁抗氧化及破骨細胞分化之抑制作用

Antioxidant Activity and Inhibitory Effect on Differentiation of Osteoclast of Onion Drink

摘要


骨質疏鬆症是盛行於全世界的退化性疾病,其中骨密度降低被認為是和氧化壓力上升與血漿中的氧化程度升高相關;而洋蔥被認為具有調節骨質代謝之能力。 本研究擬探討洋蔥汁對改善骨質疏鬆症與抗氧化之功效,採隨機雙盲安慰劑對照之試驗,共有24位受試者參與。本試驗將受試者分為兩組,一組飲用洋蔥汁,另一組飲用安慰劑飲料;試驗前一星期為飲食調整期,試驗期為八周,試驗期後兩星期為觀察期。受試者每天餐後飲用一次100mL洋蔥汁或安慰劑。試驗期間於第0、2、6、8、10周進行體位測量和血液採集,於試驗之起始,分別對三位停經後婦女進行骨質密度檢測;且於體外試驗以巨噬細胞RAW264.7誘發為破骨細胞的模式,探討洋蔥對破骨細胞分化之影響。 結果顯示,試驗組在飲用八周後會降低鹼性磷酸酶的活性,並且增加血中總抗氧化能力以及GSH之含量。骨密度部分,過重能使停經後婦女的骨密度維持優於正常體重;且於體外試驗結果則指出洋蔥汁可降低分化過程中的破骨細胞生成率。因此證實洋蔥汁具有改善骨質疏鬆之功效。

Osteoporosis is an epidemically degenerative disease. Decreasing of bone mineral density (BMD) is associated with the elevation of oxidative stress. In the past study, onion was thought effective to bone metabolism. This study was focused on the effect of onion juice on the improvement of osteoporosis and antioxidation. This was a randomized, placebo controlled and double blind design. Twenty-four subjects were involved in two groups in this study. Each subjects intook 100mL of onion juice or placebo drink every day. One week before trial was run in period. Trial period was eight weeks and two weeks after trial were follow-up period. Anthropometric measurements and blood collection were obtained at initial, 2nd, 6th, 8th and 10th week. BMD of three postmenopausal women of subjects were measured at initial and 8th week. In vitro study, the model of macrophage cell RAW264.7 differentiation into osteoclasts was used to evaluate the effect of onion juice. Therefore, after eight weeks of intervention, use of onion juice decreased the activity of alkaline phosphatase and increased antioxidant activity and contentsof GSH in plasma. Kept of BMD was better for overweight postmenopausal women than normal body weight. In vitro study, onion juice decreased the formation of osteroclast during differentiation. In conclusion, onion juice showed effect on the improvement of osteoporosis.
