摘要


本研究主要項目為聚乙二醇-鹽水兩相之三成分系統液液相平衡數據量測與蛋白質在聚乙二醇-鹽-水單相區之溶解度量測。三成分液液相平衡數據是利用原子吸收光譜儀量測鈉離子含量以推算鹽類濃度,並利用折射儀來量測折射指數來推算PEG濃度。水溶性聚合物選用PEG6000,三種鈉鹽選取硫酸鈉、硫酸氫鈉和氯化鈉,壓力在常壓下,溫度選定在20℃、25℃、30℃。蛋白質溶解度的測量操作,以PEG6000-硫酸鈉-水為系統,蛋白質選用雞卵蛋白,壓力在常壓下,溫度選定在室溫25℃。 實驗結果顯示,含硫酸鈉、硫酸氫鈉系統之液液相平衡呈現溫度相依之效應,氯化鈉之系統則觀看不到相分離的現象;蛋白質溶解度隨PEG濃度的增加而減少。液液相平衡數據則以The Extended UNIQUAC model 作關聯計算。

The subject of the present study include the measurement of liquid-liquid equilibrium data and protein solubility in single phase of polyethylene glycol – salt – aqueous two phase system. The salt concentration was measured with Atomic Absorption spectrophotometer and the PEG concentration of liquid-liquid equilibrium data were measured with Refractometer. The polymer of glycol was PEG6000, and the salts were sodium sulfate, sodium bisulfate and sodium chloride. Experiment temperatures were chosen at 20℃, 25℃, 30℃, and in the normal pressure. The protein solubility was measured with PEG6000 – sodium sulfate – water system; protein was Albumin from chicken egg. Experiments were being in the normal pressure and at 25℃. According to experimental data, the phase behavior of sodium sulfate and sodium bisulfate system depend on temperature. Sodium chloride system can not spread to two phase. The augmentative relationship between protein solubility and PEG is reverse. All of experimental data will correlate by The Extended UNIQUAC model.
