蘇木是中藥調劑經常使用的藥材,根據先前的研究,瞭解蘇木在體外實驗模式可殺死 精蟲;所以本研究以口服與睪丸被膜下注射的方式進行動物實驗,發現1.口服劑量(2,10, 300mg)經服藥及停藥之過程後,對睪丸製精功能產生暫時可逆性的傷害,然而高劑量(600mg ) 經服藥及停藥之過程後,製精功能並沒有回復原本的功能.2. 經由睪丸被膜下注射的方 式(2,10,300mg,600mg)實驗結果則顯示不同劑量所造成了永久性不可逆的不同製精功能傷 害 在顯微鏡觀察中較特別的發現包括:(1)口服蘇木2,10,300mg濃度前後睪丸組織其睪 丸製精功能受抑制,在副睪丸內之成熟精蟲數與正常值比較明顯降低,經停藥數日( 7~28 日)其成熟精蟲數則回復正常值(2)而在大劑量600 mg口服投予下,部份精原小管發生因血 栓而導致的組織壞死. 經停藥數日( 7~28 日)其成熟精蟲數亦無法回復成正常值.(3) 睪 丸被膜下注射蘇木(2,10,300,600mg) 經停藥數日( 7~ 42 日)其成熟精蟲數則無法回復成 正常值且製造精子功能中斷,部份組織壞死至只有Sertoli Cell,其睪丸組織腫大並部份呈 現黏合的現象.顯示睪丸被膜下注射蘇木比口服投予方式更具有明顯的精子製造抑制作用, 而相對地其睪丸組織破壞作用也更加明顯. 本研究並進一步以掃瞄電子顯微鏡來觀察 睪丸、副睪丸在殺精劑的作用下所受的影響;比起一般病理切片,從掃瞄電子顯微鏡可以 看出副睪丸小管內精蟲的分佈,在受到阻塞或藥物影響之後,小管內的形態變化,而睪丸 內的製精情況也可以超顯微結構去分析;經由電子顯微鏡觀察投藥前後之睪丸及副睪切片 , 發現投藥後之副睪丸內小管腔呈現塌陷及壞死的情形,而且也無法觀察到成熟的精子.這 是在文獻上最先以電子顯微鏡掃瞄睪丸製精、副睪丸小管結構的病理研究。 本研究 是以傳統中藥-蘇木的殺精作用,在不同劑量 ,不同給藥方式的動物實驗來觀察篩檢,並做 電子顯微鏡估證這種篩檢模式可用來建立評估男性避孕藥的藥效和回復性,也可在將來用 於中藥可能有害於生育的副作用偵測關鍵字:蘇木,殺精劑,男性避孕藥.
Caesalpinia sappan L. is a popular Chinese herb. According to Prf.Chiang's pre vious screening study, it has a strong spermatocidal effect in vitro. In this study, we apply for the rabbit model with oral and intratesticular injection o f Caesalpinia sappan L.and are engaged in the observations of change about th e male reproductional system through Scanning Electrical Microscopy to evaluat e the spermatocidal effect in vivo. The preliminary result shows that (1) can impair the spermatogenesis of rabbit through oral intake or intratesticular in jection and the changes between light microscopy and Scanning Electrical Micro scopy are relatively similar.(2)The inhibitory effects of spermatogenesis can occur in the situation of application of higher dosage (300,600mg) both with o ral and intratesticular injection.(3) The breakdown and collapse or recovery f rom the period of withdrawal of Caesalpinia sappan L. while application of di fferent dosage can be observed through the analysis with Scanning Electrical M icroscopy. The model can be applied not only for the screening of male contrac eptive, but also for the detection of the side-effect concerning male infertil ity in the ordinary use of Chinese herbs.Key words: Caesalpinia Sappan, Sperma tocidal, Male Contraceptive.
