愛玉子(Ficus pumila var. awkeotsang)為桑科(Moraceae)榕屬(Ficus)之常綠灌木,主要生長在台灣的中低海拔地區,為台灣之特有亞種。愛玉子之果實常經加工後以供食用或作為飲品。在過去並未有任何化學及藥理相關之文獻,僅有其原種薜荔(Ficus pumila)之相關研究。本實驗經由活性篩選得知愛玉子枝分配萃取後的甲醇層具有抗發炎和增進HaCaT細胞(人類皮膚角質細胞)Nrf2因子活化的活性。因此,將愛玉子枝的甲醇層作為研究題材進行後續的分離和研究。 經一系列管柱層析分離純化後,共28個化合物被分離出來,其中包含一個新香豆素類化合物並命名為ficumarin (1),以及27個已知化合物2‒28。化合物之結構皆經由比對已知化合物之核磁共振圖譜與質譜等資料確定。另外將所有化合物進行誘發HaCaT細胞Nrf2轉錄因子活化之活性測試,實驗結果顯示化合物5和17具有良好之誘發活性。
Ficus pumila var. awkeotsang (Moraceae) is an evergreen scandent shrub that grows in low and middle altitude areas of Taiwan. The genus is an endemic subspecies, and the fruit of F. pumila var. awkeotsang is widely used for food or beverage in Taiwan. The chemical constituent and pharmacology of F. pumila var. awkeotsang were not investigated before, but its original species (Ficus pumila) was phytochemically investigated. Accroding to bioactivity screening of the twigs of F. pumila var. awkeotsang, we found the methanol layer showed anti-inflammatory activity and increased nuclear factor erythroid-2-related factor (Nrf2) activation of HaCaT cell. Therefore, the methanolic layer was chosen as the plant material for further study. The methanol partitioned layer of the twigs of F. pumila var. awkeotsang were separated and purified by repeated column chromatography. In a result, a new coumarin compound, ficumarin (1), together with 27 known compounds (2‒28) were identified. The structures of all isolates were determined by analyzing their NMR spectroscopic data and Mass. The activity of all isolates in Nrf2 activation of HaCaT cells were tested and evaluated. Among them, compounds 5 and 17 are siginificantly active.
