葛根(Pueraria radix)是一種常見的植物,數千年來被當作中草藥與食物來使用,可改善發熱與酒醉等症狀。本研究採用葛根萃取物(Pueraria radix extract, PRE),進行免疫、抗氧化與抗血管新生等實驗。本次實驗共分成四組:groupI(control group)、groupII(alcohol group)、groupIII(PRE group)、groupIV(PRE+alcohol group),在連續餵食八週後,抽血進行分析生化功能、血液常規檢查、GSH、GSSG、GPx、GRx、MDA、CD4+/CD8+、IgG、IgM、VEGF、TIMP-1、VEGF、MMP-2。實驗結果顯示,GSH在groupIV有明顯的上升,p<0.05。與groupI相比較,MDA在groupII有上升的結果,在groupIII與groupIV都有下降的趨勢,然而未達統計學上的意義。與groupI相比較,CD3+、CD4+ lymphocytes在groupII有降低的結果,p<0.05,在groupIV的結果則恢復至接近groupI。在IgG方面,groupII低於groupI,groupIV亦低於groupI,p<0.05,groupIII則高於其它三組。在VEGF方面,groupII高於groupI,groupIII亦高於groupI,groupIV的結果則下降至接近groupI。綜合以上實驗結果顯示,酒精對於T lymphocytes與IgG有減少的作用,葛根萃取物(Pueraria radix extract, PRE)可能可以借由提升GSH來改善因酒精所造成的氧化傷害。酒精會促進VEGF的增加,單獨服用葛根亦會增加VEGF,然而合并服用葛根與酒精可以使VEGF降低至接近對照組。
Pueraria radix was a renowned herb traditionally used as herb or food supplement to enhance antipyretic efficiency and to recover from alcoholism or fatigue in China. We investigated the effects of Pueraria radix extract (PRE) on the immunological function antioxidant capacity and angiogenesis effect in the rats treated with alcohol. Methods:Sprague–Dawley rats were divided into four group (groupI:control;groupII:alcohol intake;groupIII:PRE;groupIV:PRE+ethanol). In groupII and groupIV, alcohol (99%, 6g/kg body weight) was orally administered once a day for 8 weeks. GroupIII and groupIV were supplemented with 1.7g/kg body weight of PRE once a day for 8 weeks. Results:The level of GSH was higher in groupIV than in groupII, p<0.05. Alcohol administration decreased the levels of CD3+ cells and CD4+ cells, p<0.05. The level of IgG was decreased in groupII than in groupI, p<0.05. However, the level of IgG was also decreased in groupIV than in groupI, p<0.05. Alcohol administration increased the levels of VEGF, p<0.05. However, the level of VEGF was also increased in groupIII than in groupI, p<0.05. Besides, the level of VEFG was lowered in groupIV than in groupIII, p<0.05, and near to the control group. Conclusion:This study showed that alcohol can decrease the amount of CD3+ and CD4+ lymphocytes and IgG, besides, alcohol can increase the level of VEGF. Puerariae radix extract (PRE) has the potential to induce angiogenesis. When combined treated with PRE and alcohol, the level of GSH was increased and the level of VEGF was decreased.
