台灣朝鮮薊鮮葉之萃取與生物活性分析探討

Extraction and Bioactivity Analysis of Taiwan Artichoke Fresh Leaves

摘要


朝鮮薊富含多種生物活性化合物其抗氧化能力已被廣泛研究。主要影響其抗 氧化能力來源為類黃酮及酚類化合物。不同部位所含的化合物含量也不一樣,在 這裡我們將朝蘚薊葉子在各種溫度且不同時間下使用純水萃取並對其進行抗氧化 分析,然後我們發現萃取時間並不會影響其抗氧化能力,但在 DPPH 和 ABTS 測 詴中,當在萃取溫度較低時,其抗氧化能力較不好,而 FRAP 測詴則是萃取溫度 較低時抗氧化能力較佳。另外我們也進行了細胞存活測詴和黑色素含量的分析。 在三種萃取溫度下 50 ppm 都有明顯的細胞毒性,100°C 水萃物在 10-20 ppm 下且 萃取時間為 20 min、80°C 水萃物在 20 ppm 下且萃取時間為 20 min、60°C 水萃物 在 20-40 ppm 下且萃取時間為 20 min 的有明顯的黑色素抑制效果。

Artichokes are rich in various bioactive compounds whose antioxidant capacity has been extensively studied. The main sources of its antioxidant capacity are flavonoids and phenolic compounds. The content of compounds contained in different parts is not the same. Here we extracted the leaves of artichoke with pure water at various temperatures and at different times and conducted antioxidant analysis on them. Then we found that the extraction time did not affect its antioxidant properties, in the DPPH and ABTS tests, when the extraction temperature is low, its antioxidant capacity is poor, while in the FRAP test, the antioxidant capacity is better when the extraction temperature is low. In addition, we also carried out cell viability test and analysis of melanin content. Cytotoxicity was observed at 50 ppm of all extracts. 10-20 ppm of the extract from 100°C and 20 min, 20 ppm of the extract from 80°C and 20 min, and 20-40 ppm of the extract from 60°C and 20 min significantly inhibit melanin biosynthesis in B16F10 cells.
