酒精脫氫酶等解酒產品在米格魯犬的解酒效力

Alcohol Dehydrogenase and Its Dealcoholization Activity in Beagle Dogs

摘要


由粉碎麵包酵母菌 (broken Saccharomyces cerevisiae ﹔BSC ) 體中抽取酒精脫氫酶 ( alcohol dehydrogenase; ADH ),並購取9種市售解酒產品,首先進行試管內測試檢測其分解酒精之效力及找出ADH 之最適當之分解酒精pH值,再以胃管給予beagle犬米酒後求測出血漿中含酒精濃度曲線,供選定犬隻給酒後之適當採血檢測酒精濃度時間。其後再將ADH、BSC和9種解酒商品分別以胃管給予beagle犬後15分鐘後,再以胃管給予含20 %酒精之米酒 ( 12.5ml/Kg ),於米酒給予後60分鐘檢測beagle犬血漿中之含酒精濃度 ( BAC ),供推算ADH、BSC及9種解酒商品分別在beagle犬給予後之解酒效力。結果發現ADH在試管試驗中以在 pH 8.5時之分解酒活性最佳, ADH ( 11070 unit/ml ) 添加於含0.14%酒精的血漿中60分鐘後可將其酒精濃度降至0.06%,然在胃酸pH值 ( 3.0 ) 時,ADH僅可將血漿中之酒精濃度於60分鐘後從0.16%降至0.12% 。在beagle犬體內試驗給予ADH (約40,000 unit/隻),BSC ( 30g/隻) 或解酒商品之解酒效力時,僅見ADH、BSC和一種解酒商品分別可20、7 和10 % 之降低BAC,其他8種解酒商品僅具3%>之降低BAC。ADH、BSC和其他解酒商品在beagle 犬之實驗僅略見解酒效力。

Alcohol dehydrogenase ( ADH ) extracted from the completely broken Saccharomyces cerevisiae (BSC) cells of the dried form baking yeast powder were evaluated of its dealcoholization activity in vitro at variant pH condition in order to figuring out of its most suitable working condition. ADH ( 11070 unit/ml ) provided the highest activity in vitro at pH 8.5 by decreasing the ethanol content from 0.14 to 0.06% in 1 hr, but only decreasing the ethanol content from 0.16 to 0.12% at pH 3.0. BSC and 9 commercial dealcoholizing products were also included in this study. Blood alcohol content ( BAC ) curve of beagle dogs evaluated at different intervals post Rice wine ( 20 % ) given by gastric tube was also then figuring out. ADH, BSC and other 9 commercial dealcoholizing products were individually given by gastric tube to beagle dogs following by given with the Rice wine ( 12.5ml/Kg ) at 15 later. BAC of each dog was evaluated at 1 hour post wine given for figuring out the dealcoholization activity of ADH, BSC as well as the 9 commercial dealcoholizing products in vivo. ADH, BSC and one commercial product was found with the slight decreasing of BAC at 20, 7 and 10 % level respectively. The other 8 products were found only decreasing of the BAC at less for 3% level. ADH, BSC and other dealcolizing products only provide slight dealcololization activity in vivo.
