不同製備方法及抗生素添加對牛富血小板血漿中生長因子濃度之影響

The Effect of Different Preparation Method and Adding Antibiotics on Growth Factors Concentration in Bovine Platelet-Rich Plasma

本研究旨在探討不同製備方法及抗生素添加對牛富血小板血漿(Platelet-rich plasma, PRP)中生長因子的影響。血小板中含有多種具有生物活性物質,其中生長因子被認為具有促進傷口癒合的作用,在臨床治療已漸被運用。然而,在牛隻中製備PRP的方案以及添加抗生素對生長因子的影響卻鮮少被研究。本試驗以臺灣黃牛為研究對象,共分為三個試驗。試驗一在探討不同的離心次數對PRP中血小板濃度的影響,比較一次離心與二次離心後,各層的血小板濃度。而試驗二在評估不同轉速和時間對血小板及生長因子濃度的影響。而試驗三為使用八種離心方案比較PRP中血小板及生長因子PDGF-BB、TGF-β1及VEGF的濃度。研究結果顯示,一次離心比二次離心更可以獲得較高的PRP的血小板濃度。在探討不同的時間及轉速的離心方案中,發現不同的離心方案對中以1,469 g/10 min有較高的血小板濃度。而大部分的抗生素及抗菌劑對生長因子無顯著影響,但Cefquinome及Ceftiofur可能會降低TGF-β1的濃度。結論為建議在製備牛PRP時,可採用一次離心法1,469 g/10 min,以提高血小板濃度。

This study investigated the effects of different preparation methods and antibiotic addition on growth factors concentration in bovine platelet-rich plasma (PRP). Platelets contain a variety of bioactive substances, among which growth factors are considered to have the effect of promoting wound healing and have been gradually used in clinical treatment. However, the preparation of PRP in cattle and the effect of adding antibiotics on growth factors have rarely been studied. Taiwan yellow cattle were used as research subjects in this experiment, which was divided into three trials. The effect of different centrifugation steps on platelet concentration in PRP was explored in Trial 1, comparing the platelet concentration in each layer after one step centrifugation and two step centrifugations. Trial 2 evaluated the effects of different speeds and steps on platelet and growth factor concentrations. Eight centrifugation protocols were used in Trial 3 to compare the concentrations of platelets and growth factors PDGF-BB, TGF-β1, and VEGF in PRP. It was shown in the results that a higher platelet concentration in PRP could be obtained with one step centrifugation than with two step centrifugations. In exploring the centrifugation protocols with different times and speeds, it was found that 1,469 g/10 min had a higher platelet concentration among different centrifugation protocols. Most antibiotics and antibacterial agents had no significant effect on growth factors, but Cefquinome and Ceftiofur may reduce the concentration of TGF-β1. The results suggest that one step at 1,469 g for 10 min is recommended for bovine PRP preparation to enhance platelet concentration.
