利用高效液相層析法來測量牙用麻藥管的微量酒精滲漏

The Measurement of Ultra Trace Levels of Alcohol Leakage in Dental Local Anesthetic Cartridges by HPLC/ED

摘要


一般牙科用的局部麻藥管常用酒精來浸泡消毒,但臨床醫師常懷疑會有酒精滲入麻藥管中,因為注射時,偶而引起患者有燒灼感之情形發生,在以往文獻中,也未曾有實驗証實有酒精滲漏。本次實驗的目的即在利用高效液相層析法/電化學偵測器(HPLC/ED)來驗測用酒精來浸泡麻藥的安全性,是否有微量的酒精滲入管中。實驗的方法為取數管使用過的及未使用過的麻藥管(Scandonest, 2%, 內含10.00 ng/ml epinephrine),分別浸泡於95%酒精(內含7.60% ng/ml dopamine)24小時、48小時、72小時。epinephrine是用做麻藥滲漏到酒精的標計,而dopamine是用做酒精滲漏到麻藥的標計。再分別利用高效液相層析法/電化學偵測器(HPLC/ED)來驗測酒精及麻藥管內的成份。結果發現24小時、48小時、72小時後,酒精中並沒有發現epinephrine的存在,麻藥管中也沒有dopamine的存在。因此,用酒精來浸泡麻藥管在72小時內是相當安全的。

Anesthetic cartridges usually should be permitted to soak in alcohol solution for sterilization, however, burning sensation was reported by some of the patients following injection of local anesthestic solution. The goal of this experiment is to detect the safety of storage of dental cartridge with alcohol by using High-performance liquid chromatography/ electrochemical detection (HPLC/ED) technique. 12 used and 12 nonused Scandonest (2% Mepivacaine, containing 10.00ng/ml Epinephrine) vials were stored in 95% alcohol (containing 7.60 ng/ml Dopamine) for various intervals (0, 24, 48, and 72 hours); Epinephrine in Scandonest and Dopamine in alcohol were used as markers to evaluate the leakage from drug vial to alcohol container or vice versa. Scandonest and alcohol were withdrawn separately and diluted to 1/100 by mobile phase at 0, 24, 48, and 72 hours after storage, then High-performance liquid chromatography/ electrochemical detection (HPLC/ED) was applied to measure the concentrations of the epinephrine and Dopamine in Scandonest and alcohol Solution, respectively. In conclusion, there is no detectable leakage during the storage of Scandonest in 95% of alcohol within 72 hours.
