薄荷醇(-)-menthol在活體試驗中會抑制老鼠體內血癌細胞的增生

摘要


雖然左旋薄荷醇(-)-menthol在體外試驗可造成血癌細胞(leukemia HL-60)的破壞,但動物試驗目前尚未被證實。本研究主要是著重在左旋薄荷醇對老鼠體內血癌細胞的增生是否有抑制作用。結果發現左旋薄荷醇在體外試驗可抑制血癌細胞的增生,且薄荷醇的濃度越高,效果越好。當BALB/c老鼠被注射血癌細胞WEHI-3至體內,並進行一週的血癌細胞誘導期,之後用1mg/Kg或10mg/Kg的薄荷醇治療3週,其造成不成熟的Mac-3與CD11b比例減少,且造成成熟的CD19比例增加,代表薄荷醇濃度無論1mg/Kg或10mg/Kg,皆具療效。經由薄荷醇治療之後,不僅肝脾腫大的現象明顯消褪,解剖組織也具改善。因此薄荷醇在活體試驗中會抑制老鼠體內血癌細胞的增生。

Although (-)-menthol had been reported to induce cell death in human leukemia HL-60 cells, there are no reports to show (-)-menthol affects leukemia cells in vivo. Therefore, the present studies are focused on the in vivo effects of (-)-menthol on murine leukemia WEHI-3 cells. (-)-menthol affecting murine WEHI-3 cells in vivo were initially examined, and the results indicated that (-)-menthol induced cytotoxicity and that this effect was dose-dependent. The effects of (-)-menthol on WEHI-3 cells in vivo (BALB/c mice) were also examined. We injected WEHI-3 cells to the animals, and then Animals were treated with 1mg/kg or 10 mg/kg (-)-menthol for up to 3 weeks. The results indicated that (-)-menthol decreased the percentage of Mac-3 and CD11b markers, indicating that the differentiation of the precursor of macrophage and T cells were inhibited. (-)-menthol also increase the percentage of CD19 markers, indicating that mature cell is enriching. Both the sizes of liver and spleen decrease after treatment. Based on pathological examination, the function of (-)-menthol was observed in the liver and spleen in the mice. (-)-menthol affects WEHI-3 cells in vivo.
