補腎養肝合劑對四氯化碳誘導急性肝損傷保護作用之影響

Protective Effect of Tonifying Kidney and Nourishing Liver Mixture on Carbon Tetrachloride-induced Acute Liver Injury in Rats

摘要


本研究目的在探討補腎養肝合劑萃取物對四氯化碳誘導大鼠急性肝損傷之影響。在四氯化碳誘導急性肝損傷模式中,四氯化碳組血清中ALT、AST活性顯著升高,口服補腎養肝合劑萃取物(100, 500mg/kg)可有效降低四氯化碳誘發之ALT、AST活性;並可提升肝臓中抗氧化酵素SOD、GPx、GR活性,結果顯示補腎養肝合劑萃取物對四氯化碳誘導急性肝損傷具有保護作用,其保護四氯化碳誘導急性肝損傷之機轉與提升肝臟內抗氧化酵素GR、GPx與SOD活性有關。 綜合以上結果顯示,補腎養肝合劑萃取物對四氯化碳誘導大鼠急性肝損傷有保護作用,其改善四氯化碳誘導大鼠急性肝損傷之作用機轉與增強抗氧化酵素SOD,GRd,GPx活性,改善肝組織學病理情況有關。

Obsjective: The purpose of this study is to investigate the hepato-protective effect and possible mechanism of the Tonifying Kidney and Nourishing Liver Mixture (TKNLM) in CCl4 induced acute liver injury in rats. Methods: The study analyzed ALT, AST activities in the serum, and the activities of the antioxidant enzymes (SOD, GPx, GR) in the liver. Results: When rats were treated with CCl4 in the absence of TKNLM, the activities of ALT and AST were increased, while the activities of the antioxidant enzymes were decreased. When the rats were treated with CCl4 in the presence of TKNLM, the activities of ALT and AST were significantly reduced and the activities of SOD, GR and GPx were remarkably increased. On histological examination, the administration of TKNLM showed preventive effect against liver necrosis and vacuolization. Conclusion: TKNLM possessed hepatoprotective effect. The hepatoprotective mechanism of TKNLM may be due to increasing the activities of the antioxidant enzymes (SOD, GPx and GRd) in the liver.
