何氏養巢顆粒干預DOR小鼠卵巢功能的藥效學研究

Pharmacodynamic studies of He's Yangchao Decoction on Ovarian Function in DOR Rats

摘要


目的:研究何氏養巢顆粒對小鼠卵巢儲備功能下降(DOR)的防治作用。方法:取動情週期正常ICR雌鼠分為對照組、模型組、DHEA組及養巢顆粒低、中、高劑量組,每組各10只,除對照組外其他組均採用環磷醯胺(CTX)誘導DOR小鼠模型,分別予生理鹽水及脫氫表雄酮(DHEA)、養巢顆粒灌胃8周。陰道塗片觀察動情週期,分時段檢測血清促卵泡刺激素(FSH)、促黃體生成素(LH)、雌二醇(E2)、抗穆勒氏管激素(AMH)、抑制素B(INHB),解剖後計算卵巢、子宮係數,光鏡下觀察卵巢形態組織學變化。結果:與對照組比較,模型組小鼠動情週期延長,FSH、LH值升高且AMH下降(P<0.05);與模型組比較,DHEA組、養巢顆粒各劑量組的FSH值下降,E2、INHB值升高,高劑量組AMH顯著增高(P<0.05);相較於第4周,第8周模型組及DHEA組的FSH值較前明顯升高(P<0.05),而養巢顆粒組小鼠的FSH、LH較前次檢測無明顯變化(P>0.05)。觀察卵巢組織形態可見相較於模型組,DHEA及養巢顆粒各組均可見卵巢中各級卵泡數目增多且閉鎖卵泡減少。結論:何氏養巢顆粒能夠調節性激素水準、減緩卵泡閉鎖,對改善卵巢儲備功能具有良好的效果,能夠及時防治DOR的進一步發展。

OBJECTIVE: To study the preventive and therapeutic effects of He's Yangchao Decoction on ovaries of rats with Diminished Ovarian Reserve (DOR). METHODS: ICR female rats with normal estrus cycle were divided into control group, model group, dehydroepiandrosterone (DHEA) group, and low-, medium-, and high-dose groups of Yangchao Decoction, with 10 rats in each group. All other groups except the control group were treated with cyclophosphamide (CTX). Saline, DHEA (11mg.kg-1), low dose (2.4g.kg-1), medium dose (4.8g.kg-1) and high dose (9.6g.kg-1) of Yangchao Decoction was administered for 8 weeks. Vaginal smear was used to observe estrous cycle. Two times of serum follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E2), anti-Mullerian hormone (AMH), and inhibinB were detected. The ovarian and uterus coefficients were calculated after dissection, and the morphology of ovary and the growth and development of follicles were observed under light microscope. RESULTS: Compared with the control group, the estrous cycle of the model group was prolonged, the FSH and LH values were increased, and the AMH was decreased (P<0.05). Compared with the model group, the FSH values of the DHEA group and the Yangchao Decoction decreased, E2 and inhibin B (INHB) value increased, the LH value of the medium and high dose Yangchao Decoction group decreased, and the AMH of the high dose group increased significantly (P<0.05); The FSH values of the model group and the DHEA group were significantly higher than before (P<0.05), while the FSH and LH values of the Yangchao Decoction group had no significant change compared with the previous one (P>0.05). The morphology of ovarian tissue was observed, and compared with the model group, the number of follicles in the ovary increased and the number of atretic follicles decreased in the DHEA group and the Yangchao Decoction group. CONCLUSION: He's Yangchao Decoction can regulate the level of sex hormones, slow down follicular atresia, and has a good effect on improving ovarian reserve function, and can prevent and control the further development of DOR.
