台灣育成的花椰菜F1商業栽培品種及其親本間遺傳歧異度之研究

Studies on Genetic Variation among Commercial Cultivars and Parental Lines of Cauliflower in Taiwan

摘要


本試驗在分析國內自行育成之花椰菜品種之親本之親緣關係及適合做F1純度鑑定之分子標誌。參試品種(系)包括16個為F1品種及其32個親本的自交系和2個固定品種。利用120個逢機引子(Operon A, B, C, D, E, and L kits)進行花椰菜品種(系)間多型性篩選,篩選出OPA-1,OPA-13,OPA-18,OPB-5,OPB-15,OPE-15及OPL-18等7個可產生多型性DNA條帶的逢機引子;再進一步針對國內3家花椰菜育種公司所提供的F1品種及其親本自交系篩選試驗中,總計選獲92個多型性條帶,可以很明顯的把這3家公司品種分辨出來,而對同一家公司內大部分親本其親緣關係比較接近,不同公司則相似度較低,由此可知國內花椰菜育種公司,所使用花椰菜親本皆為其培育獨有之自交系。本研究亦找到至少30個以上只出現在F1及父本上,而不出現在母本上的分子標誌,這些標誌不但可以種原間親緣關係之探討,更有機會發展應用作為花椰菜F1品種的種子純度鑑定。

This study was to investigate genetic variation of cauliflower (Brassica oleracea var. botrytis), using 16 commercial hybrid cultivars developed in Taiwan and their parental lines and two open pollinated lines; and to find molecular markers which can be used for testing F1 seed parity. Seven oligomers from initial screening were selected and used to generate 92 polymorphic molecular markers for studying phylogenetic relationships among these 34 cauliflower Cultivars/lines from three seed companies. The phylogenetic tree of the 34 cauliflower Cultivars/lines showed that seed source was an important factory affecting genetic relationships of cauliflower, Cultivars/lines from same company were more closely related then Cultivars/lines from different companies. Moreover, more than 30 molecular markers were present in F1 hybrids and male parent but were absent in female parent. These molecular markers can be used to test the purity of F1 seeds of cauliflower and, are of potential for commercial production of hybrid cultivars.
