豬霍亂沙氏桿菌產生之細胞毒素‘P65'之定性:(I)P65對ICR小鼠毒性作用之組織病理學研究

摘要


自罹患全身性敗血症型沙氏桿菌症之病豬的肺、肺門淋巴結、脾、肝及膽囊可分離到豬霍亂沙氏桿茵(Salmonella choleraesuis),其血清型為C1;6,7;C;1,5(Serogroup;O-Ag;phase-Ⅰ Ag;phase-Ⅱ Ag),自上述豬霍亂沙氏桿菌之培養之上層中,可分離並純化一種胜肽,其分子量為65kDa(簡稱P65)。P65對小鼠巨噬細胞株(P388-D1)會產生細胞變性及壞死病變,故稱P65為細胞毒素。以P65靜脈注射ICR小鼠能引起明顯的巨脾,巨肝及肺炎病變,顯示P65對小鼠具有毒性,其所引起之組織病理變化為:肺間隔壁內有極明顯的巨噬球浸潤,肺泡內亦有少許的脫落細胞等,即呈現明顯的間質性肺炎,脾臟有局部性壞死且有明顯的巨噬細胞增生,肝臟呈多發性壞死病變,以網狀內皮細胞(RE細胞)之浸潤及增生為主徵。以瓊膠免疫擴散法試驗結果,P65與S. choleraesuis之內毒素(Endotoxin)或溶解物(Lysate)之抗原性迥然不同。

Salmonella choleraesuis were isolated from lungs, hilus lymph node of lungs, spleen, liver, and gallbladder of pigs naturally-infected with systemic septicemic salmonellosis. The isolates of S. choleraesuis were serotyped as C1; 6, 7; C; 1, 5 (Serogroup; O-Ag; phase-Ⅰ Ag; phase-Ⅱ Ag). A cytotoxin, a polypeptide with a molecular weight of 65kDa (designated as P65) could be isolated from the culture supernatant of S. choleraesuis. In vitro, P65 was cytotoxic to the cells of mouse macrophage cell line (P388-D1) by causing degeneration and necrosis. In vivo, P65 caused hepatomegaly, splenomegaly and pneumonia in ICR mice following intravenous injection at a dose of 40 μg-60 μg per mouse. Histopathological changes included a distinguished infiltration of macrophages in the alveolar septa and a few detached cells in the aveolar spaces in the lungs indicative of a typical interstitial pneumonia; multifocal necrosis and marked proliferation of macrophages in the liver and spleen. There was a distinct precipitation line between P65 and its specific antibody and partial identity between the antibody and lysate of killed S. choleraesuis by agar gel immunodiffusion test (AGID test). It indicated that the antigenicity of P65 was different from those of somatic antigens of the same S. choleraesuis. Thus, P65 may play an important role in the pathogenesis of salmonellosis.
