豬萎縮性鼻炎巴氏桿菌A型與D型菌毒素之致病性與生物性狀

Characterization of Toxin Produced by Pasteurella Multocida Serotype A and D from Atrophic Rhinitis of Pigs

摘要


由野外萎縮性鼻炎患豬分離的巴氏桿茵D型與A型菌菌株製備粗製毒素(Crude Pasteurella multocida toxin; PMTcd),研討其致病性與物理、化學及生物性狀。實驗結果顯示A型與D型產毒株製備的PMTcd在小白鼠致死試驗、天竺鼠皮膚壞死試驗、豬之攻毒試驗、以及對非洲綠猿腎細胞(Vero cell)細胞株之細胞病變效應(CPE)等並無差異。小白鼠腹腔接種試驗發現大部份在攻毒從2-3日死亡,最低致死劑量約為1.37μg。LD50試驗發現小白鼠隨攻毒劑量增加,脾臟萎縮之情形愈顯著,鏡下可見淋巴小結變小,淋巴球數目顯著減少。對天竺鼠之最小引致皮膚壞死劑量約為0.27μg/0.1 mL,而皮膚壞死力價均約為2,048倍/0.1 mL。對豬則125μg之劑量可引致中等度到重度的鼻甲骨萎縮病變。以0.014μg/0.1 mL劑量即可引致Vero cell之CPE,Vero titer均高達20,4801倍/0.1 mL,顯示Vero cell對此PMTcd毒素之高敏感性。此毒素可經56℃,30分鐘及經Trypsin、Formalin、Glutaraldehyde等處理而不活化,置於-20℃及-70℃可保存6個月無毒力下降情形。

The high virulent strains of Pesterurella multocida type A and D isolated form field cases of swine atrophic rhinitis were selected for preparing the crude toxin (PMTcd) in order to study its pathogenicity and biological natures. The results indicated that the PMTcd produced by type A or D was toxic to the tested animals causing lethality in mice (LD50 = 1.37 μg), dermonecrotic change in guinea pigs (minimal dose = 0.27 μg/0.1 mL) and atrophic nasal turbinate in pigs (> 125 μg). The PMTcd (0.014 μg / 0.1 mL) was high toxic to vero cell to form CPE. Using vero cell line to test for PMTcd was proven more sensitive than others (vero titer up to 20480 X). The PMTcd was inactivated by 56℃ 30 min and by several treatments as trypsin, formalin or glutaraldehyde. The toxic level of PMTcd was stable after 6 month s stocked at -20℃ and -70℃.
