以聚合酶連鎖反應鑑別環頸雉球蟲株與雞球蟲株之DNA指紋

Study on the Differentiation of DNA Fingerprint between the Strains of Coccidia from Ring-necked Pheasant and Chicken by Using Polymerase Chain Reaction

摘要


由高麗環頸雉(Phasianus colchicus karpowi)小腸上段及盲腸段分離到兩株球蟲,經芽胞化後鑑定為Eimeria屬球蟲。將此兩株球蟲與已鑑定之雞球蟲Eimeria acervulina及Eimeria tenella以隨機增殖多形性DNA眾合酶連鎖反應、(random amplified polymorphic DNA polymerase chain reaction; RAPD-PCR),進行DNA指紋鑑定,並以瓊膠電泳分析其指紋圖譜,結果4株球蟲具有明顯不同之DNA指紋,顯示其為不同種之球蟲。

Two isolates of coccidia were identifed as Eimeria sp. from the intestine and cecum of the ring-necked pheasant. The DNA fingerprint from these two isolates of coccidia from ring-necked pheasant were compared with Eimeria acervulina and Eimeria tenella by using RAPD-PCR (random amplified polymorphic DNA-polymerase chain reaction) method. The resulting data showed differences between coccidia of pheasant and coccidia of chicken in the electrophoretic profiles when analysed with RAPD-PCR in terms of four different species of coccidia being identified.
