Characterization and Morphologic Analysis of Bovine Serum Albumin-loaded Poly (S. C. -lactide-co-glycolide) Microspheres

摘要


在疫苗的製作上,一向以抗原搭配不同的佐劑為主,但這種劑型只有刺激免疫的功用,而無控制釋放的效果。為了開發生物可吸收性且具控制釋放的疫苗劑型,我們以poly(S. C.-Iactide-co-glycolide)(PLG)為基質,以牛血清白蛋白為抗原,利用溶劑揮發的技街,進行水/油/水劑型之微球體的製作及包裝,以研究此一劑型釋放蛋白質的模式。在經位相差顯微鏡及掃描式電子顯微鏡解析後,發現這些微球體皆呈球狀,表面相當光滑,兼具單一性(monolithic)及儲存性(reservoir)的型式,其大小平均為51.52±28.82μm。試管內釋放試驗的結果,顯示微球體具有控制釋放的特性,亦即在第二天會大量釋放牛血清白蛋白,之後會持續釋放達18天以上。綜合上述,以PLG為材質,確實可以製作具控制釋放特性的微球體,而這些結果也提供未來開發疫苗劑型的另一方向。

In order to develop a biodegradable microsphere, we prepared the poly (S.C .-Iactide-co-glycolide) (PLG) microspheres containing bovine serum album in (BSA) as a model antigen by a water-in-oil-in-water emulsion solvent evaporation method. The encapsulation efficiency of BSA in microspheres was 37±3 % and the microspheres showed a mean diameter of 51.52 ± 28.82μm. They were spherical and exhibited both monolithic and reservoir forms. In vitro release study of BSA-Ioaded microspheres displayed a controlled release kinetics with a burst effect during the first two days and a sustained release for the next 18days. This study indicates that the PLG-based microspheres could serve as a formulation for controlled-release vaccine.
