取出懷孕第9.5天之大鼠胚胎培養於36℃,擺動速率為30 rpm之恆溫養贅箱48小時,其後評估胚胎生長發育情形、形態及培養液之酸鹼值。結果顯示,免賴得各供試濃度對卵黃囊長度、胎體頭部至臀部長度、頭部長度、發育等級、種節數、死亡或萎縮比率及胎體外觀異常比率具明顯作用,免賴得誘發大鼠第9.5天胎體異常現象有小眼、無眼、前腦發育不全、頭部扁平、前腦發泡、頸部或軀幹壞死、軸體異常旋轉、軀幹短而鈍及共泄外翻、背脊與後腦嚴重分裂、前腦與中腦分裂、及中腦與後腦分裂等。並對胎體頭部至臀部長度、頸部長度、發育等級及體節數具濃度-反應關係,必芬諾則在各評估項目與對照組無顯著差異。由此可知,生體外快速致畸胎性評估方法具有鑑識輔助之功用。
In order to set up the in vitro teratology for pesticides, we cultured the whole embryos of pregnant day 9.5 rats at 360C and 30 rpm incubator for 48 hours and compared the benomyl with bifenox in developmental score, morphology, and medium pH. Abnormalities induced by benomyl were microphthalmia, anophthalmia, prosencephalic hypoplasia, flat head, prosencephaiic blisters, head or trunk necrosis, abnormal axial rotation, short and blunt trunk, cloacal extrophy, dorsally trunk and hind brain splited, fore brain and trunk splited, mid-hind brain splited. The results showed that yolk sac diameter, crown-rump length, head length, developmental score, somite number, dead or atrophy and abnormal rate were treatment-related in benomyl but not in bifenox. Based on the consistency of results in vivo and in vitro for benomyl and bifenox we concluded that in vitro assessment for teratology served as a screen of the teratologic potential of pesticides in rats.
