改良培養液製備方式對豬肺炎黴漿菌抗原之影響

Effect of the Modified Liquid Medium on Antigens from the Cultures of Mycoplasma hyopneumoniae

摘要


本研究主旨是探討使用含過濾與未過濾之商品化豬血清的Friis培養液,對Mycoplasma hyopneumoniae 抗原培養的影響。結果發現使用合未過濾豬血清之Friis培養液所培養的M. hyopneumoniae 抗原,不但具備操作簡單快速,大幅降依生產成本的優點外,同時獲得提高一倍量的抗原產量,而且含未過濾豬血清之培養液於培養過程的pH值降低幅度明顯較緩和,由穿透式電子顯微鏡的觀察結果顯示,以含未過濾豬血清之培養液所培養的抗原,其細胞結構明顯呈現較實質化的特性,而兩者所得抗原的蛋白質組成,以聚丙烯醯電泳及西方氏轉漬法分析並無顯著性差異,同時由豬隻模式的動物試驗結果顯示,使用合未過濾豬血清之培養液所培養的抗原,製成鋁膠疫苗後,經豬隻兔疫試驗結果證明引起顯著較高的抗體力價反應。因此若以製造疫苗為目的,使用合未過濾豬血清之培養法培養M. hyopneumoniae 抗原,是個可行且具備多項優點的一種方式。

This investigation aimed to compare the effect of Friis media, which comprised filtrated porcine serum or non-filtrated porcine serum on the M. hyopneumoniae antigen. The Friis medium that comprised non-filtrated porcine serum was found easier, faster and cheaper to prepare. On the other hand, it was found possible to collect double the M. hyopneumoniae cells from the non-filtrated broth than from the filtrated broth. During the incubation, the pH of the culture broth with non-filtrated porcine serum decreased smoother than that of broth with filtrated serum. No significant differences existed between the cell proteins from both media by applying electrophoresis analysis and western blotting. However, transmission electron microscopy produced more substantial M. hyopneumoniae cells from the non-filtrated broth. Moreover, in vivo experimental results further confirmed that the alumium hydroxide gel vaccine of M. hyopneumoniae, prepared from non-filtrated broth induced a solid specific immune response in pigs.
