以反應曲面法探討超臨界二氧化碳萃取肖楠精油最適化條件

Optimization for the Supercritical Carbon Dioxide Extraction of Essential Oils from Calocedrus Formosana by Response Surface Methodology

摘要


本研究利用反應曲面法,建立超臨界CO_2萃取肖楠精油之最適化技術,首先進行單因子試驗,並根據萃取率,確定壓力(30~38MPa)、萃取溫度(45~55℃)及時間(1.2~2.0hr)三項因子的複因子詴驗,再以Box-Benhken中央合成設計3變數、3層級的組合,並以萃取率為響應值,藉由Design-Expert軟體,以反應曲面法分析各項自變量及其交互作用對萃取率之影響,建立二次多項回歸方程的預測模型。藉由變方分析,結果顯示壓力、溫度以及時間三者對萃取率均呈正相關,三項參數中,以萃取壓力最具影響,其次為溫度、時間。應用反應曲面法解析,以超臨界CO2萃取肖楠精油之最適條件,萃取壓力為38.6MPa、萃取溫度為51℃、萃取時間為1.4hr,在此條件下,估算萃取率為1.38%,而萃取率之實測值與預測值二者之相關係數為0.989。

Response surface methodology (RSM) was used to optimize the supercritical carbon dioxide (SC-CO_2) extraction of essential oil from ground Calocedrus formosana. The extraction yields were firstly evaluated by single factor test, a three factors and three levels, namely. extracting pressure (30~38 MPa), temperature (45~55℃) and extraction time (1.2~2.0 hr) for the multiple testing. Utilizing the Box-Benhken central composite design (CCD) in order to optimize the parameters and the yield of C. formosana oil was used as the responsive values. The RSM was employed to analyze the results of experiments and optimized by the Design-Expert. A second-degree polynomial regression equation reflecting the relationship between the extraction rate and the above mentioned extraction parameters was established and the optimal extraction process conditions were obtained by RSM analysis. ANOVA (analysis of variance) showed that all three factors had a positive correlation, the pressure affected the extraction yields most significantly, followed by the temperature and extraction time. Application of the RSM analysis showed that optimal operational parameters for C. formosana were: extraction pressure was 38.6 MPa, extraction temperature was 51℃, extraction time was 1.4 hr. Under above mentioned optimum condition, the estimated extraction yield was 1.38%. The result was verified and the correlation coefficient between the measured value and predicted value was 0.989.
