以回應曲面法探討溫度、pH值和氯化鈉含量對Aeromonas hydrophila DNA group 1生長之影響

摘要


本研究利用回應曲面法探討Aeromonas hydrophila DNA group 1之生長影響因子,以其ATP含量表示該菌之活菌量,影響因子則包括起始pH值(4.6, 5.3, 6.3, 7.3, 8.0),培養溫度(22, 25, 30, 35, 38℃)和氣化鈉含量(0.8, 1.5, 2.5, 3.5, 4.2%)。結果發現該菌其培養之起始pH值對ATP含量影響並不大,而培養溫度和氯化鈉含量之影響則較為顯著。由結果得知該菌在下述條件時其活菌量最多(卽ATP含量最高):當培養24小時,培養溫度20.3℃~24.5℃,起始pH值6.2~6.3,氣化鈉含量0.80~0.88%;培養48小時,培養溫度25.8~27.4℃,起始pH值5.8~6.2,氣化鈉含量0.87~0.88%。

The combined effects of temperature (22, 25, 30, 35 and 38℃), Sodium Chloride Content (0.8, 1.5, 2.5, 3.5 and 4.2%), and initinal pH (4.6, 5.3, 6.3, 7.3 and 8.0) on the growth of A. hydrophila DNA group 1 were studied in tryptic soy broth using response surface model. We use ATP measurement as a biomass indicator. The results showed that the bacterial ATP content was not significantly affected by the initial pH value, while the temperature and sodium chloride content were markedly affected The optimal growth conditions were found to be temperature 20.3-24.5℃, initial pH 6.2-6.3, sodium chloride content 0.80-0.88% after incubated 24 hours; temperature 25.8-27.4℃, initial pH 5.8-6.2, sodium chloride content 0.87-0.88% after 48 hours of cultivation.
