培養基組成及培養密度對黃根節蘭種子發芽與小苗生育之影響

Effects of Medium Composition and Cultural Density on Seed Germination and Seedling Growth of Calanthe sieboldii

摘要


本研究之目的在建立黃根節蘭(Calanthe sieboldii Decne)實生苗大量繁殖系統,探討培養基組成分及培養密度對黃根節蘭種子發芽及小苗生育之影響。結果顯示,以授粉後110天的種子,培養於基本培養基3 g L^(-1)花寶1號組合20 g L^(-1)蔗糖、2 g L^(-1)活性炭及8 g L^(-1)洋菜,其發芽率為14.1%。另補充7種有機物,除了馬鈴薯泥外,其它6種添加物皆可促進發芽,其中以150 ml L^(-1)椰子水處理發芽率較佳,達39.1%。實生苗生長培養基組成以花寶1號補充15 g L^(-1)蔗糖、1 g L^(-1)活性炭及2.5 g L^(-1)水晶洋菜,在15-30株/瓶培養密度,苗生長勢及鮮重遠比花寶2號配方為佳。前者配方苗生長勢及鮮重隨培養密度增加而降低。在低密度下另添加有機物對苗生長勢未明顯改善,但添加香蕉泥則可顯著提高鮮重。在20-30株/瓶培養密度時,補充香蕉泥或馬鈴薯泥皆可明顯改善苗生長勢及鮮重。

To establish seedling propagation system, this research examined the influence of medium composition and cultural density on seed germination and seedling growth of Calanthe sieboldii Decne. The results of the germination rate showed seeds 110 days after pollination cultured on basal medium 3 g L^(-1) Hyponex No. 1, 20 g L^(-1) sucrose, 2 g L^(-1) activated carbon and 8 g L^(-1) agar was 14.1%. Six organic additives, except potatoes, could enhance seed germination. The germination rate at 39.1% was highest supplied with 150 ml L^(-1) coconut milk. The growth vigor and fresh weight of seedlings growth on the medium supplemented with Hyponex No. 1, 15 g L^(-1) sucrose, 1 g L^(-1) activated carbon and 2.5 g L^(-1) gelrite in the 15-30 plants/flask cultural density was higher than on Hyponex No. 2 basal medium, but decreased when the cultural density increased. In low cultural density, the growth vigor of seedlings was not significantly improved by organic additives, except that addition of banana increased fresh weight. In 20-30 plants/flask cultural density, addition of banana or potato significantly improved seedling growth vigor and fresh weight of seedlings.
