培養基組成分對台灣風蘭及厚葉風蘭種子發芽與小苗生育之影響

Effects of Medium Composition on in vitro Seed Germination and Seedling Growth of Thrixspermum formosanum and Thrixspermum sublatum

摘要


為大量繁殖及保育台灣原生蘭科植物風鈴屬物種-台灣風蘭及厚葉風蘭,本試驗探討培養基組成分對其種子無菌播種的效用。結果顯示,台灣風蘭及厚葉風蘭無菌播種培養基以3 g L^(-1) 花寶1 號添加50 g L^(-1) 馬鈴薯泥、20 g L^(-1) 蔗糖、1 g L^(-1) 活性炭及8 gL^(-1) 洋菜的種子發芽率佳,分別為72.8%及99.9%,且小苗生育良好。種子發芽後小苗繼代培養基,台灣風蘭以3 g L(-1) 花寶1 號添加25 g L^(-1) 香蕉泥、150 g L^(-1) 馬鈴薯泥、15 g L^(-1) 蔗糖、1 g L^(-1) 活性炭及8 g L^(-1) 洋菜在株高及植株鮮重增加量較高;厚葉風蘭則以3 g L^(-1) 花寶1 號添加100 g L^(-1) 香蕉泥、100 g L^(-1) 馬鈴薯泥、20 g L^(-1) 蔗糖、1 g L^(-1)活性炭及8 g L^(-1) 洋菜在株高、葉幅、總葉片數、葉面積、根數及植株鮮重增加量等性狀較高。

This study was carried out to explore the effect of medium composition on in vitro seed germination and growth of Thrixspermum formosanum and Thrixspermum sublatum. The results showed that the medium supplemented with 3 g L^(-1) Hyponex No. 1, 50 g L^(-1) potato, 20 g L^(-1) sucrose, 1 g L^(-1) activated carbon and 8 g L^(-1) agar gave the highest germination rate (72.8 and 99.9%) for Thrixspermum formosanum and Thrixspermum sublatum., respectively. After two round of subculture the seedlings were transferred to medium supplemented with 3 g L^(-1) Hyponex No. 1, 25 g L^(-1) banana, 150 g L^(-1) potato, 15 g L^(-1) sucrose, 1 g L^(-1) activated carbon and 8 g L^(-1) agar and this gave the best results as referred to the plant height and average plant weight on Thrixspermum formosanum. After two round of subculture the seedlings were transferred to medium supplemented with 3 g L^(-1) Hyponex No. 1, 100 g L^(-1) banana, 100 g L^(-1) potato, 20 g L^(-1) sucrose, 1 g L^(-1) activated carbon and 8 g L^(-1) agar and this gave the best results as referred to the plant height, leaf span, total number of leaf, area of leaf, number of root and average plant weight on Thrixspermum sublatum.
