高劑量與低劑量Cyclophosphamide治療不同生長時間之S-180皮下腫瘤的療效比較

摘要


ICR小白鼠皮下接種肉廇─180,約4─5天後,即可觸摸到腫瘤。長廇動物平均存活期3個月左右。腫瘤接種同時或第10天,腹腔注射一次低劑量cyclophosphamide(CY,50 mg/kg)則腫瘤生長緩慢,各有4/10及3/10的動物腫瘤消失,其餘則繼續生長,而且再次以高劑量CY(200mg/kg)治潦時並無抗藥性產生。腫瘤接種後第20天以後,低劑量CY治療無效。高劑量CY(20mg/kg)治療組,於腫瘤細胞接種初期(0天及10天組)能完全抑制或排斥腫瘤。第20天,30天及40天治療者,所有動物的腫瘤治療後的20天內均有萎縮的現象,各有6/9,4/10及3/10的動物腫瘤被排斥,其餘動物的腫瘤於20天後又開始生長。本實驗的結果顯示,S─180皮下腫瘤接種後越早治療越好;高劑量比低劑量CY療效好,這種現象可能與S─180腫瘤的低免疲性有關。

ICR mice were inoculated with 1×107 S-180 tumor cells subcutaneously. The tumor growths were palpable four to five days after inoculation. When mice were treated with a single i.p. injection of low dosage cyclophosphamide (CY, 50 mg/kg) at day zero and day ten after inoculation of tumor cells, 4/10 and 3/10 of the animals were cured. No with 20 or 30-day old tumors was cured by a low dosage of CY administration. The progressed tumor grew more rapidly than the control group after drug administration, and no drug-resistance occurred when they were treated by a second high dosage of CY. When single high dosage of CY (200 mg/kg) was administered at day zero the tumor growth was overall inhibited, the grown tumor was also completely eradicated if the drug was injected on day ten. If animals were treated with a high dosage of CY at day 20, 30 and 40 after inoculation of tumor cells, 6/9, 4/10 and 3/10 of the animals were cured respectively. No animal with either a 20 or 30-day old tumor was cured by a low dosage of CY nor were any with 50-day old tumor cured by a high dosage of CY. The results indicate that the high dosage of CY is more effective than the low dosage of CY for treatment of S-180 subcutaneous solid tumors. This suggests that tumor immunity in the animal bearing S-180 tumor is weak and incapable of rejecting the tumor even enhanced by a low dosage of CY. On the contrary, the high dosage of CY treatment potentiates the tumor immunity of the animals, but only half of the treated animals rejected the tumor after the tumoricidal activity of the CY.
