Inhibitory Effects of Isoquinoline - Type Alkaloids on Leukemic Cell Growth and Macromolecule Biosynthesis

異喹啉類生物鹼對白血病細胞生長及巨型分子生合成之抑制效果

本論文係研究由台灣產植物分離之34種異哈喹類生物鹼,即benzyltetrahydroisoguino-lines.或其N-oxides, aporphines, oxoaphorph-ines以及1-N, N-dimethylaminoethylphe-nanthrenes或其N-oxides對抑制去氧核苷,想苷及氨基酸等分別組合為DNA,RNA及protein之效價加以評估。並研究其對老鼠白血病細胞L 1210以及兩種人體白血病細胞CCRF-CEM及HL-60之生長抑制活性。發現(±)-N-methylc-oclaurine(3),(-)-norannuradhapurine HBr (19), oxoglaucine (23), dicentrine methane (27)及1-(N-methyl-N-hydroxy-aminoethyl)-3,4-methylenedioxy-7-methoxyphenanth-rene (28)具有小於10μM濃度之IC50。

A number of benzylisoquinoline alkaloids of plants indigenous to Taiwan were isolated and purified and their chemical structures were deter-mined. These compounds, namely benzyltetrahy droisoquinolines or their N-oxides, aporphines, oxoaporphines and 1-N, N-dimethylaminoethylphe-nanthrenes or their N-oxides, were studied for their potency in inhibiting precursor incorporation into DNA, RNA and protein. Inhibition of murine leukemic L1210 and human leukemic CCRF-CEM and HL-60 cell growth was also examined. Of the compounds evaluated fro cell growth inhibition, (±)-N-methylcoclaurine(3), (-)-norannuradhapurine HBr(19), oxoglaucine (23), dicentrine methane (27) and 1-(N-methyl-N-hydroxyaminoethyl)-3, 4-methylenedioxy-7-methoxyphenanthrene(28) showed an IC50<10μM.
