Prevalence of Transposons Encoding Kanamycin, Ampicillin and Trimethoprim Resistance in Isolates from Urinary Tract Infections Detected Using DNA Probes

利用去氧核糖核酸探針分析攜帶 Kanamycin, Ampicillin及Trimethoprim抗藥基因之轉移遺傳因子泌尿道感染茵株之分離率

摘要


本研究是從泌尿道感染患者分離革蘭氏陰性桿菌,並利用colony hybridization的方法分析對kanamycin, ampicillin或trimethoprim呈抗性的菌株中,轉移遺傳因子transposon Tn5, Tn3, Tn7各別存在的頻率。 轉移遺傳因子Tn5,攜帶kanamycin抗藥基因,當取HindⅢ限制酶切取的片斷做探針(pro-be)來進行(colony hybridization)時,發現60株對kanamycin呈抗藥性菌株中,有38.2%呈陽性反應。此外探討129株對ampicillin呈抗藥性菌株中Tn3存在的情形。若取限制酶SacI-BamHI切取所得的3.3 Kb片斷做探針時,則有42.6%呈陽性反應。若以BamHI限制每切取所得之1Kb片斷做為探針,以探討75株對trimethoprim具藥菌株中Tn7出現的機率。結果發現有52%於colony hybridization實驗中呈陽性反應。 Southern hybridization更進一步顯示受檢臨床菌株之抗藥基因存在於細菌質體上。

Drug resistant Gram-negative bacteria causing urinary tract infections were collected. Kanamycin, ampicillin or trimethoprim-resistant strains were analyzed separately for the presence of Tn5, Tn3, or Tn7 by colony hybridization. Of these isolates, Kanamycin-resistant transposons were present in 38.2% of 60 Kanamycin-resistant isolates. A 3.3kb fragment containing SacI-BamHI transposase of Tn3 and 42.6% showed a positive reaction in 129 ampicillin -resistant clinical isolates. Among the 75 trimethoprim-resistant isolates studied, 52% were shown to contain Tn7 when probed with a 1kb BamHI fragment of Tn7. Results from Southern hybridizations demonstrated that these antibiotic resistant genes had been born on plasmids in some clinical isolates.
