Effects of Perphenazine, Hydroxyzine, and Quinidine on the Isolated Ventricular Strips of the Guinea Pig before and after Ouabain

哇巴因(Ouabain)2-氯-10{3-[4-(β-羥乙基)六氫吡嗪]丙基}苯噻嗪(Perphenazine),1-(對—氯二苯甲基)-4-[2-(L—羥乙氧)乙基]六氫吡嗪(Hydroxyzine),與奎尼丁(Quinidine)對豚鼠離體心肌條之作用

摘要


用豚鼠離體心肌做成條形標本,測定哇巴因,PERPHENAZINE,HYDROXYZINE與奎尼丁,對其基本機能之影響,結果如下: 醣苷劑如哇巴因(1:500,000),與抗心律不齊劑,如PERPHENAZINE(1:100,000),HYDROXYZINE(1:100,000),及奎尼丁(1:250,000)對心肌收縮能(Contractility),應激能(Excitability)與有效乏應期(Effective refractory period)之作用相反。用哇巴因處理後之離體心肌條收縮能,可由PERPHENAZINE與奎尼丁減弱到具有統計上意義之度。離體心肌條,先用PERPHENAZINE,HYDROXYZINE或奎尼丁處理時,哇巴因亦拮抗前者之作用,其中以對PERPHENAZINE與奎尼丁,有統計上意義。無論先用哇巴因,再用抗心律不齊劑,或先用抗心律不齊劑再試驗哇巴因,心肌條之應激能,不呈重要之改變。對離體心肌條,在哇巴因處理後,PERPHENAZINE則延長其有效乏應期,其改變統計上有意義。如先用抗心律不齊劑處理,再試驗哇巴因,其有效乏應期則更延長,在HYDROXYZINE與奎尼丁處理下者,具統計上之意義。

The effect of ouabain, perphenazine, hydroxyzine, and quinidine on the contractility, excitability, and effective refractory period of the isolated right ventricular strips was studied in the guinea pig. It was found that ouabain (1:500,000) caused an increase of contractility and excitability and a decrease of the effective refractory period, perphenazine (1:100,000), hydroxyzine (1:100,000) and quinidine (1:250,00) each caused a decrease of contractility and excitability and an increase of the effective refractility and effective refractory period was reversed by perphenazine, hydroxyzine, and quinidine, whereas in strips conditioned with perphenazine, hydroxyzine, or quinidine, the effect on contractility only of these drugs was reversed by ouabain, while their effect on the effective refractory period persisted. In fact the effective refractory period was further prolonged after ouabain. In strips conditioned with one of the drugs no consistent changes in excitability were observed follwing any of the testing drug(s).
