Some Newer Antihypertensive Drugs on Coronary Blood Flow

數種抗高血壓新藥對冠狀血流量之影響

摘要


同時記錄血壓,心臟排出量與心動電流圖,以研究Guanethidine及Bretylium與作用短暫之Sodium nitroprusside,Azamethonium,Chlorisondamine,Pentolinium以及Trimethaphan對犬冠狀血流量之作用。並推算外圍總組力與冠狀阻力。在十四個實驗中,一個作對照,三個研究Guanethidine另三個考察Bretylium,另有七個試用於作用短暫的藥物。Bretylium之作用,在三至四小時內大部消失,而Guanethidine之作用,則持續較久。作用短暫藥物之作用,在數分鐘或半小時內,完全或部份消失。作用不論久暫,心臟排出量皆減少,而外圍總阻力與冠狀阻力,則罕呈變化。在藥物效用減血壓時,冠狀血流量減少。

The effect on the coronary blood flow of guanethidine and bretylium, and among the short-acting hypotensives, of sodium nitroprusside, azamethonium, chlorisondamine, pentolinium, and trimethaphan was studied in dogs together with simultaneous registration of the blood pressure, cardiac output and ECG. Total peripheral(TPR)and coronary resistance(CR)was calculated. Of fourteen successful experiments one was used as control, three were on guanethidine, three on bretylium and seven on the short-acting drugs. It was found that in the dose range used the effect of bretylium was largely dissipated in three tofour hours, while that of guanethidine persisted much longer. Withina few minutes to half an hour total or partial recovery occurred from the effects of the short-acting drugs. In all instances there was areduction of the cardiac output with little change in TPR and CR during the hypotensive phase. Coronary blood flow decreased during the druginduced hypotension.
