Effect of Medium Nitrogen Source on the Growth of Tetrahymena pyriformis MC

培養液氮源對纖毛蟲生長之影響

摘要


從纖毛蟲(Tetrahymena Pyriformis MC)體內之核酸(RNA),去氧核酸(DNA),核酸/去氧核酸比,麩氨酸-丙酯酸及麩氨酸-草醋酸轉氨酶,與氨之排泄量,觀察不同類別之氮源(蛋白質或氨基酸)對於牠們生長過程中之影響,則見在不同培養氮源中所得之結果各異。酪蛋白最易被纖毛蟲吸收並用以合成其體內之蛋白質。在脨培養液中,纖毛蟲體內之核酸,轉氨酶及氨之排泄量,在培養後均降低,顯示從培養液中攝氮趨低。於氨基酸培養液中,不能證明纖毛蟲攝取培養液中氮趨低,而其緩慢之繁殖及大量氨之排泄,反映纖毛蟲所攝之氨基酸,大部份均耗於產能,而不參加蛋白質合成。

The influence of different types of nitrogen source on Tetrahymena pyriformis MC was studied during the period of growing phases of the organism. The changes of RNA, DNA, RNA/DNA ratio, glutamic-pyruvic and glutamic-oxaloacetic transaminase activities, and ammonia accumulation were determined and compared at different periods of incubation. Results indicated that casein was readily taken up and utilized by the ciliates for growth and body protein synthesis. Indications of reduced assimilation of medium nitrogen was however shown in peptone medium at later period of incubation by lower nucleic acid contents, transaminase activity and ammonia production. In amino acid medium, no evidence of a limited uptake of medium nitrogen was demonstrated. The low population and high ammonia accumulation indicated that a large proportion of assimilated amino acids were used as energy source instead of protein synthesis.
