細花野牽牛簇葉病之光學及電子顯微鏡觀察

Light and Electron Microscopic Observation of Ipomoea obscura Witches' Broom Disease

摘要


細花野牽牛簇葉病病株及感染該病之長春草病株之葉脈組織經超薄切片與電子顯微鏡觀察結果,在其病組織之篩管細胞內發現多形性擬菌質體(Mycoplasma-like organism)之存在,擬菌質體直徑約爲60~740 nm,在長春草病組織篩管細胞內較爲充滿擬菌質體。另以該長春草病組織徒手切片之Dienes' stain染色與光學顯微鏡觀察結果,在其篩管細胞內顯現專一性藍色反應,此項反應表示該篩管細胞中擬菌質體之存在。另在供試健全植株者則無此項反應亦無擬菌質體之存在。依此項擬菌質體之形態存在位置及對Dienes' stain之特殊反應與其他已知簇葉病所發現之擬菌質體相類似,該擬菌質體可能爲新記載之細花野牽牛簇葉病之病原。

Electron microscopic examinations made on thin-sections of diseased leaves from Ipomoea obscura Ker. and periwinkle infected with Ipomoea obscura witches' broom revealed the presence of pleomorphic bodies of Mycoplasma-like organisms (MLO) in the sieve cells of phloem tissues. These MLOs were found in various sizes and concentrations, and the size ranged from 60 to 740nm in diameter, and more aggregation or high concentrations in periwinkle diseased phloem tissues. Light microscopic observation made on hand-sections of diseased leaves of periwinkle showed distinctly specific blue staining reaction with Dienes' stain in sieve cells, which indicated the presence of MLOs in the phloem tissues, and gave no reaction in phloem tissues and none of MLO was found from those sections of healthy plants. Since the morphology and localization and the specific reaction with Dienes' stain of these MLOs were similar to those found previously elsewhere in witches' broom diseases, these pleomorphic bodies seemed to be the causal organism of the new recorded Ipomoea obscura witches' broom disease.
